Product Description
Flutamide-d7 is the deuterium labeled form of Flutamide used as an internal standard for the quantification of flutamide by GC- or LC-MS. Flutamide is an androgen receptor antagonist and prodrug form of 2-hydroxy flutamide with anticancer activity. Flutamide is converted to 2-hydroxy flutamide by the cytochrome P450 (CYP) isoform CYP1A2 in human liver microsomes. It is cytotoxic to PC3 and LNCaP prostate cancer cells with IC50 values of 98.8 and 81.8 µM, respectively. Flutamide (50 mg/kg per day) reduces tumor growth in a PC-82 mouse xenograft model.
Flutamide-d7 is the deuterium-labeled form of flutamide, a nonsteroidal antiandrogen used for prostate cancer; the d7 label provides a stable-isotope internal standard for LC-MS/MS quantification. Flutamide is a prodrug that is rapidly metabolized to 2-hydroxyflutamide, which competitively antagonizes the androgen receptor (AR), blocking testosterone and dihydrotestosterone binding and inhibiting AR-mediated gene transcription and prostate cancer cell proliferation.
Biological Activity I Assay Protocols (From Reference)
Targets
The molecular target is the androgen receptor (AR), where the active metabolite 2-hydroxyflutamide binds competitively and inhibits AR-mediated transcription; flutamide and its metabolites also inhibit AR nuclear translocation and DNA binding, and at higher concentrations they may interact with GABA_A receptors, but AR antagonism is the principal mechanism relevant to prostate cancer therapy.
ln Vitro
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a cause for concern [1].
In vitro, flutamide-d7 is metabolized to 2-hydroxyflutamide-d7, which competes with [3H]R1881 for AR binding, inhibits AR-responsive luciferase reporter activity in prostate cancer cells, and suppresses androgen-dependent cell proliferation; the deuterated form retains AR antagonist activity while enabling LC-MS/MS quantification of flutamide and its active metabolite.
ln Vivo
In vivo, oral flutamide-d7 is rapidly converted to 2-hydroxyflutamide, which reduces prostate tumor growth in AR-dependent xenograft models, lowers serum prostate-specific antigen, and produces antiandrogenic effects in androgen-sensitive tissues; the labeled form is used to quantify plasma and tumor concentrations of flutamide and 2-hydroxyflutamide and to relate AR occupancy to antitumor efficacy.
Enzyme Assay
Noncellular AR binding assays incubate recombinant AR ligand-binding domain or LNCaP cell lysates with [3H]R1881 and increasing concentrations of flutamide-d7 or its 2-hydroxy metabolite; bound and free radioligand are separated by hydroxyapatite or filtration, and IC50 values are derived, with AR-mediated reporter gene activity assessed in parallel.
Cell Assay
Cellular assays use LNCaP or VCaP prostate cancer cells treated with flutamide-d7; AR nuclear translocation is assessed by immunofluorescence, AR-responsive reporter gene activity by luciferase assay, and cell proliferation by MTT or BrdU incorporation, with AR expression confirmed by immunoblotting.
Animal Protocol
Animal protocols administer flutamide-d7 orally to mice bearing LNCaP or VCaP xenografts at doses of 10–100 mg/kg; tumor volume, serum PSA, and AR target gene expression in tumor tissue are measured, and plasma and tumor concentrations of flutamide and 2-hydroxyflutamide are quantified by LC-MS/MS to relate AR occupancy to antitumor efficacy.
