Product Description
Fluorescein-O-acetate is a fluorescent reagent.
Fluorescein-O-acetate (FOA) is a fluorescent labeling reagent composed of fluorescein linked to an acetate group; upon intracellular hydrolysis by esterases, it releases fluorescein, which emits bright fluorescence, and it is used for cell tracking, viability assessment, and enzyme activity measurement in fluorescence microscopy and flow cytometry.
Biological Activity I Assay Protocols (From Reference)
Targets
There is no pharmacological receptor target; the acetate group is hydrolyzed by intracellular esterases to release fluorescein, which is retained in cells with intact membranes and reports cell viability, membrane integrity, and esterase activity. The fluorescein moiety can also be activated to react with amines for isomerically homogeneous fluorescent labeling of biomolecules.
ln Vitro
In vitro, fluorescein-O-acetate is taken up by cells and hydrolyzed to fluorescein, producing bright fluorescence that allows assessment of cellular uptake and viability; it is used in cell proliferation and cytotoxicity assays, and as a fluorescent label for detecting and quantifying chemical reactions, with applications in fluorescence microscopy, flow cytometry, and ophthalmologic diagnostics.
ln Vivo
In vivo, fluorescein-O-acetate is used as a diagnostic tool in ophthalmology for fluorescein angiography to visualize blood vessels and tissues; it is also used for cell tracking in animal models, where labeled cells are detected by fluorescence imaging, and its hydrolysis product fluorescein is cleared renally, enabling visualization of vascular and renal function.
Enzyme Assay
Fluorescein-O-acetate is characterized by spectrophotometry (excitation/emission maxima of fluorescein, ~490/520 nm) and HPLC for purity; esterase activity is measured by incubating the substrate with cell lysates or purified esterases and monitoring the increase in fluorescence at 520 nm, and the rate of hydrolysis is quantified against a fluorescein standard curve.
Cell Assay
Cells are incubated with fluorescein-O-acetate (typically 0.1–10 µM) for 15–60 min, washed, and analyzed by fluorescence microscopy or flow cytometry; intracellular fluorescein fluorescence reports esterase activity and membrane integrity, and cell viability is quantified by comparing fluorescence intensity with untreated controls.
Animal Protocol
Mice or rats receive fluorescein-O-acetate by intravenous or local administration for fluorescein angiography or cell tracking; vascular and tissue fluorescence is imaged by fundus angiography or whole-body fluorescence imaging, and plasma clearance and renal excretion of fluorescein are measured by fluorescence spectroscopy of blood and urine samples.

Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 2.5619 mL 12.8093 mL 25.6187 mL
5 mM 0.5124 mL 2.5619 mL 5.1237 mL
10 mM 0.2562 mL 1.2809 mL 2.5619 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly.
