Biological Activity I Assay Protocols (From Reference)
Targets
FFN511 targets neuronal vesicular monoamine transporter 2 (VMAT2), a protein responsible for transporting monoamines including dopamine, serotonin, and norepinephrine into synaptic vesicles. By inhibiting serotonin binding to VMAT2 with IC50 = 1 µM, FFN511 serves as a fluorescent probe for studying monoamine transmission. It is taken up by VMAT2 into synaptic vesicles and released during exocytosis, allowing visualization of dopamine release.
ln Vitro
In cultivated mouse chromaffin cells, FFN511 (350 nM; 30 min) accumulates in a pattern comparable with LDCV, and the lipophilic base chloroquine eliminates this accumulation [1].
In vitro, FFN511 inhibits serotonin binding to VMAT2-containing membranes with IC50 = 1 µM. It is used as a fluorescent tracer to visualize dopamine release from individual presynaptic terminals using time-lapse microscopy. It can directly reflect release dynamics during exocytosis. These in vitro activities make FFN511 a valuable tool for studying synaptic transmission and monoamine release.
ln Vivo

Enzyme Assay
In vitro binding assays for FFN511 measure inhibition of serotonin binding to VMAT2-containing membranes. Membranes are incubated with radiolabeled serotonin and varying concentrations of FFN511. Bound radioactivity is measured, and IC50 = 1 µM is determined.

Experimental Results: Accumulation in cultured mouse chromaffin cells (same as LDCV), and accumulation can be eliminated by the lipophilic base chloroquine , thereby collapsing the vesicle pH gradient.
