Ethyl Violet (CAS: 2390-59-2) is a cationic triarylmethane dye. It is structurally recognized as the tetraethyl homolog of Crystal Violet. It is used as a stain for pancreatic tissue and elastin, in the detection of gram-negative bacteria, and as a reagent for the extractive-spectrophotometric determination of anionic surfactants. It is also a non-carcinogenic, Proposition-65-compliant alternative to Crystal Violet. The molecular weight is 492.14, formula C31H42ClN3.

Targets
Ethyl Violet is a dye and does not have a specific biological target. It stains biological tissues by binding to negatively charged molecules like nucleic acids and proteins through ionic and hydrophobic interactions. It is used as a pH indicator, changing from yellow at pH 0.0 to blue at pH 3.5.
ln Vitro
Ethyl Violet is used in vitro as a staining reagent. It is employed in histology to stain pancreatic tissue and elastin. It is also used in microbiology for the detection of gram-negative bacteria. In analytical chemistry, it is used for the extractive-spectrophotometric determination of anionic surfactants.
ln Vivo
Ethyl Violet is not used in vivo for therapeutic or diagnostic purposes. Its applications are limited to in vitro staining and analytical chemistry.
Enzyme Assay
For staining, tissue sections or bacterial smears are incubated with an Ethyl Violet solution. The stain binds to the target structures, providing color contrast for microscopic examination. For analytical applications, it is used in a liquid-liquid extraction followed by spectrophotometric measurement.
Cell Assay
In cell-based assays, Ethyl Violet is used as a staining agent. Cells are fixed and then incubated with the dye. The stained cells can be observed under a microscope to visualize cellular structures. It is also used in viability staining in some contexts.
