Ingredients for the EthD-1 Working Solution and Methods of Use 1.1 Make a stock solution of EthD-1 by mixing 2 mM DMSO with it. Note: It is advised to aliquot the EthD-1 storage solution and keep it in the dark at -20°C or -80°C. 1.2 Components of the working solution: To create a 0.1–10 μM EthD-1 working solution, mix the storage solution with either PBS or previously prepared serum-free cell culture media. Note: Please prepare the EthD-1 working solution for immediate usage by adjusting its concentration to suit the current circumstances. Staining 2.1 Cells Centrifugation is used to gather the cells, then PBS is added and the cells are washed twice for five minutes each time. Adherent cells: take out the trash cells and put in the digested islet cells. Add PBS and wash twice for five minutes after centrifugation and discarding the supernatant. Note: Slide staining can be used to stain adhering cells in the absence of flow cytometry investigations. 2.2 For 15 to 60 minutes, add 1 mL of the EthD-1 working solution and the stainless steel working solution. 2.3 Centrifuge for 3–4 seconds at 4°C at 400 g. 2.4 After adding PBS, wash the cells twice for five minutes each. 2.5 Re-suspend the cells in 1 milliliter of PBS or serum-free water, and examine using a flow cytometer or fluorescence microscope. Observations 1. After aliquoting, it is advised to store the EthD-1 storage solution in the dark at -20°C or -80°C to prevent recurrent freezing and thawing. It can be kept for one month at -20°C and for six months at -80°C. 2. Please modify the EthD-1 working fluid concentration based on the current circumstances. 3. This product may not be used for clinical diagnosis or treatment, nor may it be included into food or medication. It is intended solely for professional use in scientific study. 4. Please wear disposable gloves and a lab coat for your health and safety.
