In rabbit infused with δ-Sleep Inducing Peptide (DSIP), the spindle activity is increased, and heart and respiration rate are moderately decreased. δ-Sleep Inducing Peptide (DSIP) may act as a programming modulator at supra-operational level rather than as a transmitter at operational level[1]. δ-Sleep Inducing Peptide (DSIP; 40 μg/kg, i.p.) increases catalase and superoxide dismutase (SOD) activities and malonic dialdehyde (MDA) concentration in the liver homogenate of rats subjected to acute stress, but the opposite effects are observed at 120, and 360 μg/kg
Emideltide (CAS 62568-57-4), also known as Delta Sleep-Inducing Peptide (DSIP), is a synthetic nonapeptide with the amino acid sequence H-Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu-OH and molecular weight of 848.81 g/mol. It is a neuropeptide that induces delta sleep in mammals. Emideltide has antioxidant and anxiolytic properties. It has been studied for its effects on sleep, stress, and circadian rhythms.

Emideltide targets neuropeptide systems involved in sleep regulation and stress response. ln Vitro
In vitro, emideltide has antioxidant properties. Its effects on neuronal cells and neurotransmitter systems have been studied. The peptide may modulate oxidative stress and neuroinflammation in cellular models. Its ability to induce sleep and reduce anxiety has been demonstrated in animal studies. However, detailed in vitro activity data is limited.
ln Vivo
In rabbits, delta-sleep-inducing peptide (DSIP) produces decreased heart and respiratory rate control and increased spindle activity [1]. Delta-sleep-inducing peptide (DSIP; 40-360 μg/kg, i.p.) rapidly labels homogenates at 40 μg/kg malondialdehyde (MDA) concentration and boosts catalase and superoxide dismutase (SOD) activity in the matrix [2]. However, the correct effect was found at 120 μg/kg and 360 μg/kg.
In vivo, emideltide (DSIP) produces decreased heart and respiratory rate and increased spindle activity in rabbits. It induces delta sleep in mammals. It has anxiolytic effects in animal models. Plasma DSIP concentrations are correlated with the human circadian rhythm and are dependent on the initiation of sleep. The peptide may act as a suprachiasmatic programming modulator.
Enzyme Assay
In vitro receptor binding assays for emideltide are not well-established, as its specific receptor has not been definitively identified. Binding studies may use brain membrane preparations and radiolabeled DSIP or related peptides. Functional assays measure effects on neuronal activity in brain slices or cultured neurons. Antioxidant activity is assessed using cell-free or cell-based ROS detection assays.
Cell Assay
Cellular assays for emideltide use neuronal cell cultures or brain slices. Cells are treated with various concentrations of emideltide. Neuronal activity is assessed by electrophysiological recordings or calcium imaging. Oxidative stress markers are measured using fluorescent dyes. Neurotransmitter release is measured using HPLC or microdialysis.
Animal Protocol
In vivo animal studies for emideltide utilize rabbits, mice, or other mammals. The peptide is administered via intracerebroventricular (ICV) injection or intravenous infusion. Sleep-wake patterns are monitored by EEG/EMG recordings. Heart and respiratory rates are measured. Anxiety-like behavior is assessed using elevated plus maze or other behavioral tests. Circadian rhythm parameters are measured.
ADME/Pharmacokinetics
Emideltide has molecular weight of 848.81 g/mol and is a nonapeptide. As a peptide, it has poor oral bioavailability and is typically administered via injection or central routes for research purposes. Pharmacokinetic properties include rapid clearance by peptidases. The peptide's short half-life necessitates frequent dosing or continuous administration in vivo studies. It is stored as a powder at -20°C.
Toxicity/Toxicokinetics
Preclinical toxicity of emideltide is not extensively documented. As an endogenous neuropeptide, it is generally considered to have low toxicity. In animal studies, the peptide is well-tolerated at the doses used for sleep and anxiety research. No significant adverse effects have been reported. Standard toxicology studies would be required for therapeutic development.
