| Targets |
Isotope-labeled phosphatidylcholine; no intrinsic receptor or enzyme agonist/antagonist activity. Endogenous DPPC is a major lung surfactant phospholipid and a structural component of cell membranes and liposomes; its biological roles are mediated by membrane organization and surface tension reduction. The deuterated standard is used as an analytical surrogate for DPPC quantification and does not modulate phospholipase or surfactant protein activity.

| ln Vitro |
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a matter for concern [3].
The d66-DPPC standard shows no measurable intrinsic bioactivity and is not used for potency screening. The high deuterium content provides a mass-shift signal that is well separated from endogenous DPPC and other phospholipids; the standard corrects for extraction recovery, ionization efficiency, and matrix effects during DPPC quantification in surfactant, liposomal formulations, and biological matrices. |
| ln Vivo |
No intrinsic in vivo pharmacological activity has been reported for the deuterated standard. DPPC-d66 is used as an isotope-dilution internal standard to quantify endogenous DPPC in lung tissue, bronchoalveolar lavage fluid, and liposomal formulations, and to track the biodistribution of DPPC-containing delivery systems.

| Enzyme Assay |
LC-MS/MS DPPC quantification: add DPPC-d66 to the sample before lipid extraction; extract phospholipids with chloroform/methanol; separate on a reversed-phase C18 column; monitor the specific m/z transition of the d66 standard (e.g., [M+H]+ at m/z 184 for the phosphocholine headgroup) and the corresponding unlabeled transition. Quantification is by isotope-dilution calibration using peak-area ratios.
|
| Cell Assay |
Cell-based DPPC analysis: incubate cells with DPPC-containing liposomes or surfactant preparations, wash, lyse, extract lipids, and quantify DPPC by LC-MS/MS using DPPC-d66 as internal standard.
|
| Animal Protocol |
In vivo lung surfactant and biodistribution analysis: collect bronchoalveolar lavage fluid or lung tissue, add DPPC-d66 before extraction, isolate phospholipids, and quantify DPPC by LC-MS/MS to assess surfactant composition and liposome biodistribution.

Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). Injection Formulation 4: DMSO : 20% SBE-β-CD in saline = 10 : 90 [i.e. 100 μL DMSO → 900 μL (20% SBE-β-CD in saline)] Oral Formulations
Oral Formulation 1: Suspend in 0.5% CMC Na (carboxymethylcellulose sodium) Oral Formulation 2: Suspend in 0.5% Carboxymethyl cellulose Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
