Product Description
Donepezil-d5 is the deuterium labelled form of Donepezil. Donepezil (E2020 free base) is an AChE inhibitor (antagonist) with IC50s of 8.12 nM and 11.6 nM for bovine AChE and human AChE, respectively.
Donepezil-d5 (CAS 1128086-25-8) is the deuterium-labeled form of Donepezil (E2020 free base), a specific and potent acetylcholinesterase (AChE) inhibitor. Donepezil is used for the treatment of Alzheimer's disease. Donepezil inhibits bovine AChE and human AChE with IC₅₀ values of 8.12 nM and 11.6 nM, respectively. The deuterated compound is used as an internal standard for the quantification of Donepezil by mass spectrometry. It has a molecular formula of C₂₄H₂₄D₅NO₃ and a molecular weight of 384.52.
Biological Activity I Assay Protocols (From Reference)
Targets
Donepezil-d5 targets acetylcholinesterase (AChE), the enzyme responsible for the hydrolysis of the neurotransmitter acetylcholine in the synaptic cleft. Donepezil is a specific and potent AChE inhibitor with IC₅₀ values of 8.12 nM and 11.6 nM for bovine AChE and human AChE, respectively. By inhibiting AChE, Donepezil increases acetylcholine levels, improving cognitive function in Alzheimer's disease.
ln Vitro
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a cause for concern [1].
In vitro, Donepezil-d5 is used as a tracer to study the AChE inhibition and pharmacokinetics of Donepezil. The compound does not exhibit intrinsic pharmacological activity beyond that of Donepezil but serves as a quantitative tool for studying AChE inhibition, drug metabolism, and neuropharmacology in cell-based systems. Donepezil inhibits AChE with IC₅₀ values of 8.12 nM and 11.6 nM for bovine and human AChE.
ln Vivo
In vivo, Donepezil-d5 is used in pharmacokinetic and pharmacodynamic studies to trace the distribution and metabolism of Donepezil in animal models. The deuterium label allows for precise quantification of the drug and its metabolites in plasma and tissues using mass spectrometry. The compound is used in Alzheimer's disease research.
Enzyme Assay
As an internal standard, Donepezil-d5 is used in analytical assays such as LC-MS/MS. Typical protocols involve spiking the labeled compound into biological samples prior to extraction and analysis. The labeled compound co-elutes with the unlabeled Donepezil but is detected at a different mass-to-charge ratio, allowing for precise quantification and correction for matrix effects.
Cell Assay
In vitro cell-based experiments using Donepezil-d5 involve treating neuronal cells with the compound and analyzing its uptake and metabolism by mass spectrometry. The compound can be used to study AChE inhibition, cholinergic signaling, and the pharmacokinetics of Donepezil in various cell types.
Animal Protocol
In vivo animal studies using Donepezil-d5 involve administering the compound to rodents via oral gavage. Blood samples are collected at various time points, and the concentration of the labeled compound is measured by mass spectrometry. Tissue distribution and metabolic profiling can also be performed.
ADME/Pharmacokinetics
Donepezil-d5 exhibits pharmacokinetic properties similar to those of unlabeled Donepezil. Donepezil is well-absorbed after oral administration and has good brain penetration. It is metabolized in the liver by CYP enzymes and excreted primarily in urine. The deuterium label provides a distinct mass shift for analytical detection without significantly altering the compound's physicochemical properties.
Toxicity/Toxicokinetics
Donepezil-d5 is considered safe for research use at typical concentrations. Unlabeled Donepezil has a well-established safety profile as an approved Alzheimer's disease therapeutic but can cause adverse effects such as gastrointestinal disturbances. As a stable isotope-labeled compound, it is not intended for therapeutic use and is handled under standard laboratory safety practices.
