| Targets |
No classical pharmacological receptor or enzyme target. The compound is a fluorescent dye; its “target” is the detection of fluorescence or the preparation of labeled biomolecules after derivatization. It does not modulate biological activity.

| ln Vitro |
Advice (This is our suggested protocol, which should be adjusted to suit your particular circumstances as it simply offers guidance). Labeling of cells: 1. HepG2 cells are cultured at 37°C in a 5% CO2 atmosphere in DMEM/HIGH GLUCOSE medium, which contains 10% FBS, 100 U/mL penicillin G, and 100 μg/mL streptomycin sulfate. 2. Fill the confocal culture dish with 1×106 logarithmic half-lived HepG2 cells. For 36 hours, cells were incubated at 37°C with 5% CO2. 3. Put 200 μL of Hoechst fluorescent dye 33342 (10 μg/mL) in there. For 20 minutes, incubate the cells at 37°C with 5% CO2. 4. Put 200 μL of Cy3.5-loaded FA-PEG-RH nanomicelles in place of the culture medium. 5. After one hour, four hours, eight hours, and sixteen hours, respectively, take photos of the cells under fluorescence excitation at 350 nm and 550 nm using a fluorescence microscope.
The compound is not a biological modulator. Its in vitro “activity” is optical: it provides bright, photostable orange-red fluorescence with a high quantum yield, enabling sensitive detection in fluorescence-based assays when conjugated to a targeting moiety. |
| ln Vivo |
Direct in vivo bioactivity is not applicable. The free dye can be used for in vivo imaging only after conjugation to a targeting moiety, but the non-reactive chloride salt is primarily a synthetic precursor or fluorescence standard rather than a therapeutic agent.
|
| Enzyme Assay |
For fluorescence spectroscopy, dissolve Cyanine 3.5 chloride in aqueous buffer or DMSO and record absorption and emission spectra (ex ~581 nm, em ~596 nm). Determine the fluorescence quantum yield using a standard dye (e.g., rhodamine B) for calibration.
|
| Cell Assay |
Not applicable for standard cell-based assays; the non-reactive dye is not used directly for cellular labeling unless conjugated to a targeting moiety. For membrane labeling, incubate cells with a reactive derivative (e.g., NHS ester) prepared from the free dye, then wash and image using a Cy3.5 filter set.
|
| Animal Protocol |
For in vivo imaging, conjugate a reactive derivative of Cyanine 3.5 to a targeting moiety ex vivo, purify, and administer intravenously (e.g., 1–10 mg/kg). Image at defined time points using a fluorescence imaging system compatible with 581/596 nm excitation/emission. Include free-dye and unconjugated-ligand controls.

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). Injection Formulation 4: DMSO : 20% SBE-β-CD in saline = 10 : 90 [i.e. 100 μL DMSO → 900 μL (20% SBE-β-CD in saline)] Oral Formulations
Oral Formulation 1: Suspend in 0.5% CMC Na (carboxymethylcellulose sodium) Oral Formulation 2: Suspend in 0.5% Carboxymethyl cellulose Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
