Cy5-DBCO (CAS 1564286-24-3) is a fluorescent dye conjugate containing a Cy5 fluorophore and a dibenzocyclooctyne (DBCO) group. With a molecular weight of 1148.42 g/mol and the formula C₆₄H₇₇N₅O₁₂S₂, this compound is used in bioconjugation and imaging applications. The Cy5 fluorophore has excitation and emission maxima at approximately 650 nm and 670 nm, respectively, making it suitable for near-infrared fluorescence imaging. Biological Activity I Assay Protocols (From Reference)
Targets
Cy5-DBCO targets azide-containing molecules for labeling via strain-promoted azide-alkyne cycloaddition (SPAAC). The DBCO group reacts with azides with high efficiency and selectivity, forming a stable triazole linkage. This bioorthogonal reaction does not require copper catalysis, making it suitable for labeling in living systems. The Cy5 fluorophore provides a bright and photostable signal for near-infrared fluorescence imaging, allowing for the detection and tracking of labeled biomolecules in vitro and in vivo.
ln Vitro
Without the use of a copper catalyst or high temperatures, Cy5-DBCO (DBCO-SuLfo-Cy5) combines with azide through a copper-free "click chemistry" reaction to generate a stable triazole.
In vitro, Cy5-DBCO is used to label azide-containing biomolecules, such as proteins, peptides, and nucleic acids, via SPAAC. The labeling reaction is typically performed in aqueous buffers at room temperature for 1-2 hours. The labeled biomolecules can then be analyzed by SDS-PAGE, Western blot, or fluorescence microscopy. Cy5-DBCO is also used in the synthesis of fluorescently labeled PROTACs and other conjugates for cellular imaging studies.
ln Vivo
In vivo, Cy5-DBCO is used for near-infrared fluorescence imaging of azide-labeled biomolecules. The compound's excitation and emission maxima in the near-infrared range allow for deep tissue imaging with minimal background autofluorescence. Cy5-DBCO-labeled probes can be used to track the biodistribution of drugs, study protein-protein interactions, and image tumors in animal models.
Enzyme Assay
In vitro labeling assays for Cy5-DBCO involve measuring the efficiency of the reaction with azide-containing molecules.
