Chlorthal-dimethyl (DCPA, Dacthal) is a chlorinated benzoic acid herbicide used for preemergence control of annual grasses and broadleaf weeds in various crops. It affects cell division and differentiation, interfering with the normal development of vascular tissue. It has been registered as a pesticide in the US since 1958.

Targets
Chlorthal-dimethyl acts as a plant growth regulator and herbicide. It causes localized swelling and disrupts mitosis in germinating seedlings. It interferes with cell division and differentiation, affecting the normal development of vascular tissue. Its herbicidal activity is selective, targeting specific weed species while sparing crops.
ln Vitro
In vitro, chlorthal-dimethyl affects cell division and differentiation in plant cell cultures. It causes localized swelling and disrupts mitosis in germinating millet seedlings. It is a plant growth regulator that affects mitosis and cell division.
ln Vivo
In vivo, chlorthal-dimethyl is used for preemergence weed control in turf, ornamentals, strawberries, and agronomic crops including cotton, soybeans, and field beans. It is effective against smooth and hairy crabgrass, witchgrass, green and yellow foxtails, fall panicum, and other annual grasses.
Enzyme Assay
In vitro herbicidal activity is assessed using plant cell cultures or seedling growth assays. Seeds are germinated in agar or soil containing chlorthal-dimethyl at various concentrations. Germination rate, root elongation, and shoot growth are measured. Microscopic analysis assesses mitotic disruption and vascular tissue development.
Cell Assay
Plant cell cultures (e.g., from millet, Arabidopsis, or crop species) or whole seedling assays are used. Cells or seedlings are treated with chlorthal-dimethyl at various concentrations. Cell division is assessed by counting mitotic figures. Vascular tissue development is evaluated by histological staining and microscopy.
Animal Protocol
In vivo herbicidal efficacy is evaluated in greenhouse or field trials. Chlorthal-dimethyl is applied preemergence as a spray or granular formulation. Absorption, Distribution and Excretion
Metabolized in animals to monomethyl ester and chloral, and excreted in urine. Metabolic studies in humans and dogs have shown that… ester bond hydrolysis… leads to the excretion of monocarboxylic acid or dicarboxylic acid derivatives in urine. The remainder is excreted unchanged in feces. No significant storage in body tissues has been found. No dacalorie residues were detected in the milk of dairy cows fed this compound feed. Approximately 1% intact dacalorie was detected in feces. Dacaloic acid (2,3,5,6-tetrachloromonomethylterephthalic acid) is primarily excreted via feces (3.3%) and urine (90.9%). …Not absorbed by leaves. …Not transported by plants. …Kills germinating seeds; the exact mechanism is unclear. …Not considered to be metabolized by plants.
Metabolism/Metabolites
Dacaloic acid (2,3,5,6-tetrachloromonomethylterephthalic acid) is primarily excreted in feces (3.3%) and urine (90.9%). Liver tablets rapidly produce this metabolite. Dacaloic acid does not break down when incubated with rumen contents.
Studies in humans and dogs show that ester bonds undergo significant hydrolysis…leading to the excretion of monocarboxylic acid or dicarboxylic acid derivatives in urine.

Chlorthal-dimethyl has a molecular formula of C₁₀H₆Cl₄O₄ and a molecular weight of 331.96. It has a melting point of 155-156°C. It is a chlorinated aromatic ester.
