In Fura-2-loaded neuronal cells, CALP3 TFA (50, 100, 150, and 200 μM) dose-dependently suppresses glutamate, which causes a substantial sustained rise in [Ca2+]i (IC50=37.25 μM). In cultured rat neocortical neurons, (50, 100, 150, 200 μM) reduces glutamate-induced cytotoxicity in a dose-dependent manner (IC50=50.97 μM). Apoptosis is dose-dependently inhibited by CALP3 TFA (IC50=33.41 μM)[1]. In human T cells, HIV gp120 and SAg-induced apoptosis is inhibited by 100 μM of CALP3 TFA[1]. CALP3 TFA (100 μM; 15 min pretreatment) enhances the proportion of viable cells and decreases gossypol-induced necrosis[2]. Fmoc-Asp(PEG-PS)-OAl is the starting point for the synthesis of Cyclic-CALP3. Since cyclic CALP3 cannot block Ca21 inflow, a negative control peptide was used. Glutamate's action is not inhibited by cyclic CALP3[1].
