Product Description
C24-Ceramide (d18:1/24:0) is a sphingomyelin used to form lipid membranes. C24-Ceramide (d18:1/24:0) induces time-dependent changes in membrane properties. C24-Ceramide (d18:1/24:0) induces membrane reorganization.
C24-Ceramide (d18:1/24:0) is a naturally occurring very-long-chain sphingolipid that serves as both a structural membrane component and a signaling molecule. It is the direct precursor of C24-sphingomyelin and is enriched in detergent-resistant membrane domains, where it modulates membrane order, permeability and receptor clustering. Beyond its biophysical role, C24-Ceramide has been identified as a competitive binding agonist of phosphatidylinositol 5-phosphate 4-kinase type-2 gamma (PIP4K2C), a regulator of mTOR complex signaling, linking ceramide metabolism to growth control.
Biological Activity I Assay Protocols (From Reference)
Targets
PIP4K2C (phosphatidylinositol 5-phosphate 4-kinase type-2 gamma); mTOR signaling pathway; membrane lipid organization.
ln Vitro
C24-Ceramide binds and activates PIP4K2C, promoting mTOR signaling activation in gallbladder cancer models. It induces time-dependent changes in membrane properties and membrane reorganization, increasing membrane order and permeability. As a competitive binding agonist of PIP4K2C, it drives downstream mTOR pathway activity and supports cancer cell progression, with functional effects observed in the low micromolar range in cell-based assays.
ln Vivo
In vivo, C24-Ceramide supports gallbladder cancer progression through PIP4K2C-dependent mTOR activation. Elevated C24-ceramide levels correlate with tumor growth and poor prognosis in gallbladder cancer, and pathway activation is observed in xenograft models. Its lipophilic nature requires formulation for systemic administration, and oral bioavailability has been reported for the PIP4K2C-agonist activity.
Enzyme Assay
For PIP4K2C binding assays, recombinant PIP4K2C is incubated with a fluorescent or radiolabeled phosphatidylinositol 5-phosphate probe and increasing concentrations of C24-Ceramide in assay buffer containing detergents to mimic membrane conditions. Bound ligand is separated by filtration or measured by fluorescence polarization, and Ki is derived from competition curves.
Cell Assay
For cell-based assays, gallbladder cancer cells or PIP4K2C-expressing lines are treated with C24-Ceramide (1–50 μM) for 24–72 h. mTOR pathway activation is assessed by phosphorylation of S6K1, 4E-BP1 and AKT by Western blot, and cell proliferation is measured by MTT or colony formation. Membrane order is assessed with Laurdan or di-4-ANEPPDHQ imaging.
Animal Protocol
For in vivo studies, mice bearing gallbladder cancer xenografts receive C24-Ceramide in a lipid formulation (e.g., 10–50 mg/kg, i.p. or oral) for 2–4 weeks. Tumor volume is monitored, and tumor tissue is analyzed for PIP4K2C activity, mTOR pathway phosphorylation and apoptosis by immunohistochemistry, with plasma and tissue ceramide levels quantified by LC-MS/MS.

Additional Infomation
N-Tecosylsphingosine is an N-acylsphingosine in which the N-acyl group of the ceramide is designated as a tetracosyl group. It is a mouse metabolite. It is an N-acylsphingosine, Cer(d42:1), and N-(very long chain fatty acyl)sphingosine base. It is functionally related to tetracosyl acid.
