How to apply 1. 1.1 Preparation of stock solution: Dissolve 1 milligram of BODIPY 493/503 in 382 μL DMSO to create a 10 mM stock solution. This completes the preparation of the working solution. Note: It is advised to keep the stock solution between -20°C and -80°C in storage. To obtain a 1-10 μM working solution, do not use 1.2 BODIPY 493/503 working solution without bone marrow or PBS extraction solution. Note: Please modify the working solution for BODIPY 493/503 based on the current circumstances. 2. Suspension cells (6-well plate) 2.1 Cell staining a. Discard the supernatant after centrifuging at 1000g for three to five minutes at 4°C. Use PBS to wash twice, for five minutes each. There is b cell density. For five to thirty minutes, add one milliliter of the working solution. 1×106/mL. Discard the supernatant after centrifuging at 400 g for three to four minutes at 4°C. d. Use PBS to wash twice, for five minutes each time. e. Re-suspend the cells in PBS or serum-free cells. 2.2 Adherent cells as observed using a flow cytometer or fluorescence microscopy a. On sterile coverslips, cultivate adherent cells. b. Empty the coverslip of any extra cells by removing it from the culture medium. C. To fully cover it, add 100 μL of working solution and give it a little shake. Prelude: 5–30 minutes for cells. d. Use water to wash twice, for five minutes each time. observation using flow cytometry or fluorescence microscopy. For a year, store at -20°C; shield from light. Observations 1. Kindly modify the BODIPY 493/503 working fluid concentration based on the current circumstances. 2. Prior to conducting the next studies, Saudi cells were first cultured with 30 μM oleic acid for eight hours. 3.
