Biological Activity I Assay Protocols (From Reference)
Targets
AFP-464 free base targets HIF-1α (as an inhibitor with IC50 of 0.25 μM) and the aryl hydrocarbon receptor (AhR) as a potent activator. HIF-1α is a transcription factor that regulates responses to hypoxia. AhR is a ligand-activated transcription factor involved in xenobiotic metabolism. Activation of AhR by AFP-464 leads to DNA binding and apoptosis.
ln Vitro
AFP-464 free base is an active HIF-1α inhibitor with an IC50 of 0.25 μM. It is also a potent AhR activator. AhR activation leads to phosphorylation of p53, induction of p21Waf1/Cip1, and apoptosis. Detailed in vitro data are available.
ln Vivo
AFP-464 free base is an active HIF-1α inhibitor and AhR activator. It induces apoptosis in cancer cells.
Enzyme Assay
In vitro HIF-1α inhibition assays using cell-based reporter systems or by measuring HIF-1α protein levels and transcriptional activity. Cells are treated with AFP-464 under hypoxic conditions. AhR activation assays measure AhR binding and downstream gene expression.
Cell Assay
In vitro cell-based assays using cancer cell lines. Cells are treated with AFP-464 at various concentrations. HIF-1α activity and AhR activation are assessed. Cell viability is assessed using MTT assays. Apoptosis is assessed by measuring p53 phosphorylation, p21 expression, and caspase activity.
Animal Protocol
In vivo studies in animal models of cancer. AFP-464 is administered via various routes at doses determined from pharmacokinetic studies. Tumor growth inhibition is assessed. HIF-1α and AhR target gene expression in tumors is measured.

Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 1.8364 mL 9.1821 mL 18.3641 mL
5 mM 0.3673 mL 1.8364 mL 3.6728 mL
10 mM 0.1836 mL 0.9182 mL 1.8364 mL
