Product Description
Acetoxycycloheximide is a novel and potent apoptosis inducer
Acetoxycycloheximide is an antibiotic with potent antitumor properties, primarily functioning as a protein synthesis inhibitor. It induces cell surface TNF receptor 1 and rapidly induces apoptosis mediated by the release of cytochrome c via activation of c-Jun N-terminal kinase (JNK). It also downregulates cell surface TNF-R1.
Biological Activity I Assay Protocols (From Reference)
Targets
Acetoxycycloheximide targets the protein synthesis machinery, specifically the ribosome, to inhibit translation. By inhibiting protein synthesis, it triggers apoptosis through the JNK pathway. It also modulates TNF receptor 1 levels, affecting TNF-α-mediated signaling.
ln Vitro
In vitro, Acetoxycycloheximide induces apoptosis by activating the JNK pathway and releasing cytochrome c from mitochondria. It also downregulates cell surface TNF-R1. Its activity is measured in cell viability assays, where it shows potent antitumor activity. It induces procaspase-3 activation.
ln Vivo
In vivo, Acetoxycycloheximide has been studied for its antitumor properties. Its mechanism of inducing apoptosis and inhibiting protein synthesis makes it a potential anticancer agent. Specific in vivo data are not detailed in the search results.
Enzyme Assay
The in vitro assay for Acetoxycycloheximide involves measuring its ability to inhibit protein synthesis, which can be assessed using a cell-free translation system. Its ability to induce apoptosis is measured by caspase activity assays or by assessing cytochrome c release from mitochondria.
Cell Assay
Cellular in vitro experiments for Acetoxycycloheximide are conducted using cancer cell lines. Cells are treated with the compound, and cell viability is assessed using MTT or similar assays. Apoptosis is measured using Annexin V staining or caspase activity assays. The activation of JNK and release of cytochrome c are assessed by Western blot.
Animal Protocol
In vivo animal experiments for Acetoxycycloheximide would involve using mouse xenograft models of cancer. The compound would be administered, and tumor growth would be monitored. Tumors would be harvested for analysis of apoptosis markers.

Note: Please be aware that the above formulations are for reference only. InvivoChem strongly recommends customers to read literature methods/protocols carefully before determining which formulation you should use for in vivo studies, as different compounds have different solubility properties and have to be formulated differently.

(Please use freshly prepared in vivo formulations for optimal results.)
Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 2.9465 mL 14.7327 mL 29.4655 mL
5 mM 0.5893 mL 2.9465 mL 5.8931 mL
10 mM 0.2947 mL 1.4733 mL 2.9465 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. • Click the “Calculate” button
  • The answer of 17.513 mg appears in the Mass box. In a similar way, you may calculate the volume and concentration.

Dilution Calculator allows you to calculate how to dilute a stock solution of known concentrations. For example, you may Enter C1, C2 & V2 to calculate V1, as detailed below:

What volume of a given 10 mM stock solution is required to make 25 ml of a 25 μM solution?
Using the equation C1V1 = C2V2, where C1=10 mM, C2=25 μM, V2=25 ml and V1 is the unknown:
  • Enter 10 into the Concentration (Start) box and choose the correct unit (mM)
