Oseltamivir acid-d3 is a deuterated form of Oseltamivir acid. Oseltamivir acid, the bioactive metabolite of Oseltamivir phosphate, is an orally bioactive and selective inhibitor of influenza virus neuraminidase (IC50=2 nM), active against both influenza viruses A and B.
Oseltamivir acid-d3 (CAS 1242184-43-5), also known as GS 4071-d3, Ro 64-0802-d3, or oseltamivir carboxylate-d3, is a deuterated form of oseltamivir carboxylate, the active metabolite of the neuraminidase inhibitor oseltamivir phosphate (Tamiflu). Oseltamivir acid-d3 contains three deuterium atoms replacing hydrogen atoms. The compound is used as an internal standard for analytical methods such as LC-MS/MS for the quantification of oseltamivir and its active metabolite in biological samples. Oseltamivir is an antiviral drug used for the treatment and prevention of influenza A and B.
Biological Activity I Assay Protocols (From Reference)
Targets
The primary target of oseltamivir acid-d3 is the influenza virus neuraminidase (NA) enzyme, the same target as the parent compound oseltamivir. Neuraminidase is a surface glycoprotein of influenza viruses that cleaves sialic acid residues, facilitating viral release from infected cells. Inhibition of neuraminidase prevents viral release and spread, reducing the duration and severity of influenza illness. The deuterated form has the same mechanism as the parent compound.
ln Vitro
In vitro antiviral activity of oseltamivir acid-d3 is expected to be identical to that of oseltamivir carboxylate. Oseltamivir carboxylate potently inhibits influenza virus neuraminidase, with IC₅₀ values in the low nanomolar range against influenza A and B viruses. The compound is active against both seasonal and pandemic influenza strains. Oseltamivir acid-d3 is used as an internal standard in analytical methods rather than for biological activity studies.
ln Vivo
In vivo efficacy of oseltamivir acid-d3 is not studied as a therapeutic agent; the parent prodrug oseltamivir phosphate is the clinically used formulation. Oseltamivir phosphate is well absorbed after oral administration and is rapidly converted by hepatic esterases to the active metabolite oseltamivir carboxylate. The active metabolite has a half-life of approximately 6-10 hours and is eliminated primarily by renal excretion. Specific PK parameters have been extensively characterized.
Toxicity/Toxicokinetics
The toxicological profile of oseltamivir acid-d3 is identical to that of oseltamivir. Oseltamivir has a favorable safety profile based on extensive clinical data. Common adverse effects include nausea, vomiting, and headache. Rare but serious adverse effects include neuropsychiatric events.
