Product Description
6-Maleimidocaproic acid sulfo-NHS is a PROTAC (PROteolysis TArgeting Chimera) linker of the alkyl/ether class, may be utilized to prepare PROTAC protein degraders.
6-Maleimidocaproic acid sulfo-NHS (CAS 103848-61-9) is a heterobifunctional crosslinker containing a maleimide group and a sulfo-NHS ester. Formula C14H16N2O9S, MW 388.35. The sulfo-NHS ester is water-soluble and reacts with primary amines on proteins or ligands to form stable amide bonds. The maleimide group reacts with thiol groups on cysteine residues to form stable thioether bonds. It is used for bioconjugation, ADC synthesis, and protein crosslinking with orthogonal amine and thiol reactivity.
Biological Activity I Assay Protocols (From Reference)
Targets
The compound itself has no intrinsic biological target. In bioconjugation, the sulfo-NHS ester couples to amine-containing molecules such as antibodies or proteins in aqueous buffer, while the maleimide group reacts with thiol-containing molecules such as reduced antibodies or cysteine-modified proteins. This orthogonal reactivity enables sequential attachment of two different molecules. In ADC applications, it enables conjugation of a payload to an antibody.
ln Vitro
One ligand is for an E3 ubiquitin ligase, and the other is for the target protein; these two ligands are joined by a linker to form PROTACs. To specifically destroy target proteins, PROTACs take advantage of the intracellular ubiquitin-proteasome system.[1]
In vitro activity is characterized by efficient sulfo-NHS ester amine coupling at pH 7.2-8.5 for 30-60 minutes and maleimide-thiol coupling at pH 6.5-7.5 for 2-4 hours. Conjugation efficiency is assessed by HPLC or LC-MS. The sulfo group provides water solubility, eliminating the need for organic co-solvents. The C6 spacer provides flexibility between the reactive groups.
ln Vivo
No direct in vivo biological activity is reported. Its utility arises from incorporation into conjugates or ADCs. The sulfo group's water solubility simplifies conjugation. The maleimide-thiol linkage is stable in plasma. In vivo studies typically evaluate antitumor efficacy of ADCs in xenograft models, with dosing via intravenous injection.
Enzyme Assay
For non-cellular binding assays, dissolve 6-Maleimidocaproic acid sulfo-NHS in aqueous buffer such as PBS at pH 7.2-8.0. Prepare amine-containing protein at 1-10 mg/mL and add linker at 5-10 fold molar excess. Incubate 30-60 minutes at room temperature. Quench with Tris-HCl at pH 8.0 to 50 mM for 30 minutes. Purify by desalting or dialysis. For maleimide coupling, reduce thiol-containing molecule with TCEP at 1-5 mM for 30 minutes, then add maleimide-functionalized protein at pH 6.5-7.5 and incubate 2-4 hours. Analyze by SDS-PAGE or LC-MS.
Cell Assay
For cell-based experiments, prepare the conjugate or ADC using the amine and thiol coupling protocols. Purify by size exclusion chromatography. Add to cultured cells at 1-100 nM in complete medium. Incubate 24-72 hours at 37 degrees Celsius. Assess cytotoxicity by MTT or CCK-8 assay if a payload is conjugated. Assess cellular uptake by flow cytometry or confocal microscopy if a fluorophore is attached.
Animal Protocol
For in vivo studies, formulate the conjugate or ADC in a suitable vehicle such as PBS or saline. Administer to mice via intravenous injection at 1-10 mg/kg every 2-3 days for 2-4 weeks in tumor xenograft models. Measure tumor volume twice weekly. Monitor body weight and clinical signs for toxicity. At study endpoint, excise tumors for immunohistochemistry and collect blood for pharmacokinetic analysis. All procedures follow IACUC-approved protocols.
