Product Description
C-12 NBD cholesterol is a cholesterol analogue labeled with the hydrophilic fluorophore NBD (NBD: Ex=465 nm, Em=535 nm). C-12 NBD cholesterol may be utilized to study cholesterol utilization and metabolism.
3-NBD-C12 Cholesterol is a fluorescent cholesterol analog in which a 12-carbon linker connects the cholesterol moiety to the nitrobenzoxadiazole (NBD) fluorophore (Ex/Em ≈ 465/535 nm). The NBD group is hydrophilic and environment-sensitive, enabling real-time visualization of cholesterol trafficking, membrane organization, and lipid dynamics in live cells and model membranes.
Biological Activity I Assay Protocols (From Reference)
Targets
No classical pharmacological receptor or enzyme target. The compound is a fluorescent lipid probe; its “target” is the cholesterol-rich membrane environment, where it partitions into lipid rafts and reports on cholesterol transport, esterification, and membrane organization.
ln Vitro
The compound itself is not a modulator of enzyme or receptor activity. Its in vitro “activity” is optical and lipid-based: it incorporates into membranes and reports on cholesterol distribution and dynamics by fluorescence microscopy, enabling studies of cholesterol uptake, efflux, and intracellular trafficking.
ln Vivo
In vivo data are limited. The probe is primarily used for in vitro live-cell imaging and for ex vivo tissue staining. Systemic administration for whole-animal imaging is not a standard application because of its lipophilicity and potential for non-specific membrane partitioning.
Enzyme Assay
For cell-free membrane studies, incorporate the probe into liposomes of defined lipid composition at 0.1–1 mol% and measure fluorescence emission (Ex ≈ 465 nm, Em ≈ 535 nm) to assess cholesterol-dependent membrane order.
Cell Assay
For live-cell imaging, incubate cells with 1–5 µM 3-NBD-C12 Cholesterol in serum-free medium for 15–60 min at 37°C. Wash and image by fluorescence microscopy using a GFP/FITC filter set. Include a no-probe control and a cholesterol-depleting agent (e.g., methyl-β-cyclodextrin) as a negative control.
Animal Protocol
For tissue staining, incubate fresh tissue sections with 1–5 µM probe in physiological buffer for 15–30 min at 37°C. Wash and image by fluorescence microscopy. Include a no-probe control and a cholesterol oxidase–treated sample to confirm specificity.

Note: Please be aware that the above formulations are for reference only. InvivoChem strongly recommends customers to read literature methods/protocols carefully before determining which formulation you should use for in vivo studies, as different compounds have different solubility properties and have to be formulated differently.

(Please use freshly prepared in vivo formulations for optimal results.)
Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 1.3139 mL 6.5695 mL 13.1391 mL
5 mM 0.2628 mL 1.3139 mL 2.6278 mL
10 mM 0.1314 mL 0.6570 mL 1.3139 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
