Product Description
22(R)-Hydroxycholesterol-d7 is the deuterium labelled form of 22(R)-Hydroxycholesterol.
22(R)-Hydroxycholesterol-d7 is the deuterium-labeled form of 22(R)-hydroxycholesterol (22R-OHC), an oxysterol with a hydroxyl group at the 22R position of the cholesterol side chain. 22R-OHC is a key intermediate in steroid hormone biosynthesis and a modulator of nuclear receptor signaling; it is formed by CYP11A1 and serves as a precursor to pregnenolone and other steroids, and it can modulate LXR and other nuclear receptors. The d7 label provides a mass-shift reference for isotope-dilution quantification of 22R-OHC in adrenal tissue, plasma, and steroidogenic cells.
Biological Activity I Assay Protocols (From Reference)
Targets
The primary biological target is the steroidogenic pathway and nuclear receptor signaling. 22R-OHC is a substrate for CYP11A1 in the first step of steroidogenesis and can modulate LXR and other nuclear receptors; its biological effects are mediated by enzymatic conversion to steroids and by receptor-mediated signaling. The deuterated standard is used as an analytical surrogate for 22R-OHC quantification and does not modulate steroidogenic enzymes or nuclear receptors.
ln Vitro
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a cause for concern [2].
The d7-labeled standard shows no measurable intrinsic bioactivity and is not used for potency screening. Endogenous 22R-OHC is a low-abundance oxysterol that requires a structurally matched internal standard for accurate quantification; the d7 form co-elutes with the analyte and corrects for extraction recovery, ionization efficiency, and matrix effects during oxysterol analysis.
ln Vivo
No intrinsic in vivo pharmacological activity has been reported for the deuterated standard. Endogenous 22R-OHC is a steroidogenic intermediate and a modulator of nuclear receptor signaling; the d7 standard supports in vivo quantification of 22R-OHC in adrenal tissue and plasma and enables studies of steroidogenesis and oxysterol signaling in animal models.
Enzyme Assay
LC-MS/MS oxysterol quantification: add 22(R)-Hydroxycholesterol-d7 to the sample before saponification and extraction; extract sterols with organic solvent; derivatize if required; separate on a reversed-phase C18 column; monitor the specific m/z transition of the d7 standard and the corresponding unlabeled oxysterol transition. Quantification is by isotope-dilution calibration using peak-area ratios.
Cell Assay
Cell-based steroidogenic and oxysterol profiling: culture adrenal or steroidogenic cells under conditions that modulate steroidogenesis, harvest and quench metabolism, add the d7 internal standard, extract sterols, and analyze by LC-MS/MS.
Animal Protocol
In vivo adrenal and plasma oxysterol analysis: collect adrenal tissue or plasma at defined time points, add 22(R)-Hydroxycholesterol-d7 before homogenization, saponify and extract sterols, and quantify by LC-MS/MS to assess steroidogenic flux and oxysterol levels in animal models.

5 mM 0.4882 mL 2.4408 mL 4.8816 mL
10 mM 0.2441 mL 1.2204 mL 2.4408 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
