Product Description

SRT1720 (formerly known as CAY10559) is a novel and selective SIRT1 activator with an EC50 of 0.16 μM in a cell-free assay, but is >230-fold less potent for SIRT2 and SIRT3. It shows similar activity with the known SIRT1 activator resveratrol, but is 1000-fold more potent. In animal studies it was found to improve insulin sensitivity and lower plasma glucose levels in fat, muscle and liver tissue, and increased mitochondrial and metabolic function. A study of SRT1720 conducted by the National Institute on Aging found that the drug may extend the lifespan of obese mice by 44%.

Biological Activity I Assay Protocols (From Reference)
ln Vitro
Because SRT 1720 inhibits histone acetyltransferase p300, it effectively lowers p53 acetylation in cells even when SIRT1 is not present[2].
ln Vivo
Similar to rosiglitazone treatment, SRT 1720 (10, 30, 100 mg/kg, oral) dramatically decreased hyperinsulinemia and largely restored high insulin levels after 4 weeks. Lepob/ob mice treated with SRT 1720 have considerably lower fasting blood glucose levels [1] that are almost normal. Through downstream targets of SIRT1 (such as PGC1α and FOXO1), SRT 1720 protects mice against the detrimental effects of diet-induced obesity and is linked to fatty acid metabolic adaptability and oxidative metabolism [2]. In WT mice, SRT 1720 (50–100 mg/kg, oral) lowers arterial oxygen saturation and attenuates elastase-induced air gap enlargement and pulmonary function degradation during emphysema development [3].

Solubility in Formulation 1: ≥ 2.08 mg/mL (4.43 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.8 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL.
Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.

Solubility in Formulation 2: ≥ 2.08 mg/mL (4.43 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.8 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.
