CCR4 0.02 μM (IC50)
CCR4 antagonist 4 targets the CC chemokine receptor 4 (CCR4), a G protein-coupled receptor involved in the recruitment of immune cells, particularly Th2 cells, to sites of inflammation. By blocking CCR4, this compound inhibits the signaling pathways that lead to chemotaxis and activation of these cells. |
|---|---|
| ln Vitro |
In vitro, CCR4 antagonist 4 potently inhibits CCR4 with an IC50 of 0.02 μM. It also blocks MDC-mediated chemotaxis with an IC50 of 0.007 μM and Ca2+ mobilization with an IC50 of 0.003 μM. These activities demonstrate its high potency and efficacy in blocking CCR4-mediated signaling.
|
| ln Vivo |
In vivo, CCR4 antagonist 4 is used for research into allergic inflammation. By blocking CCR4, it is expected to reduce the recruitment of Th2 cells and the subsequent inflammatory response in animal models of allergy. Specific animal model data are not detailed in the provided literature.
|
| Enzyme Assay |
In vitro enzyme or receptor binding assay protocols for CCR4 antagonist 4 typically involve radioligand binding assays to determine its affinity for CCR4. Functional assays, such as measuring MDC-induced chemotaxis or Ca2+ mobilization in cells expressing CCR4, are used to assess its antagonist activity.
|
| Cell Assay |
In vitro cell-based assay protocols for CCR4 antagonist 4 involve using cells that express CCR4, such as primary immune cells or cell lines. Cells are treated with the compound and then stimulated with a CCR4 agonist (e.g., MDC) to induce chemotaxis or calcium flux. The inhibition of these responses is measured to determine the compound's potency.
|
| Animal Protocol |
In vivo animal experimental protocols for CCR4 antagonist 4 would typically involve using mouse models of allergic inflammation, such as asthma or atopic dermatitis. The compound is administered, and endpoints such as inflammatory cell infiltration, cytokine levels, and clinical symptoms are evaluated.
|
| ADME/Pharmacokinetics |
Pharmacokinetic properties of CCR4 antagonist 4 have not been characterized in published studies. Specific parameters such as half-life, Cmax, and bioavailability have not been reported.
|
| Toxicity/Toxicokinetics |
Toxicological data for CCR4 antagonist 4 are limited. It is intended for research use only. As a research chemical, standard laboratory safety precautions should be followed.

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2.5 mg/mL (5.00 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween-80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one),clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution and add it to 400 μL PEG300 and mix well. Then add 50 μL Tween-80 to the above system and mix well. Then continue to add 450 μL of physiological saline to make up to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2.5 mg/mL (5.00 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one),clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution and add it to 900 μL of 20% SBE-β-CD saline solution and mix well. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
