In kinase activity assays, purified AMPK or SIK transfers a phosphate group from ATP to a specific serine/threonine residue within the AMARA peptide. This phosphorylation can be detected by radioactive (32P-ATP incorporation) or luminescence-based methods.

| Enzyme Assay |
In a typical in vitro kinase assay: Purified AMPK or cell lysates containing the kinase are incubated with reaction buffer (MgCl2, ATP, and 32P-ATP or unlabeled ATP) and AMARA peptide substrate (10-100 uM). After 30 min at 30degC, the reaction is stopped. Phosphorylated peptide is collected onto phosphocellulose paper or a streptavidin-coated plate (if biotinylated). Radioactivity or luminescence is measured.

| Cell Assay |
AMARA peptide is not used in live cell experiments due to its inability to cross the cell membrane. For cellular kinase activity, cells are lysed after treatment with modulators, and the lysates are then used in in vitro kinase assays with AMARA peptide to quantify AMPK or SIK activity.
