T7 tag antibody binding assay (ELISA): A microplate is coated with T7 Tag Peptide TFA (0.1-10 ug/mL) overnight at 4degC. After blocking, increasing concentrations of anti-T7 tag antibody (0-1000 ng/mL) are added and detected with an HRP-labeled secondary antibody. Alternatively, the peptide can be used as a competitor: antibody is pre-incubated with soluble peptide before adding to a plate coated with T7-tagged protein to determine IC50.

| Cell Assay |
Cell-based protein detection: Cells are transfected with a T7-tagged expression vector. After 24-48 h, cell lysates are prepared. For Western blot, the lysates are electrophoresed and transferred, then the membrane is incubated with anti-T7 tag antibody. A parallel control lane can be treated with T7 Tag Peptide TFA during antibody incubation to confirm band specificity (competitive blocking). For immunoprecipitation, the peptide can be used for elution.

Not applicable for direct in vivo use. In some models, T7-tagged proteins are administered to mice, and T7 Tag Peptide TFA can be used to block the tag for control purposes, but this is uncommon. The primary use is in vitro as a biochemical tool.
