Biological Activity I Assay Protocols (From Reference)
ln Vivo
The plasma clearance of AZD0022 in mice and dogs was 8.2 and 8.6 mL/min/kg, respectively[1]. In the GP2D model, a single oral dose of 150 mg/kg prolonged the drug exposure time in plasma and tumors. It is estimated that AZD0022 binds to tumor tissues 18 times more readily than to plasma. A dose of 150 mg/kg administered twice daily for 7 consecutive days (BID) inhibited pRSK activity by up to approximately 75%. Drug exposure-related regulation of pRSK activity was observed with doses of 10, 50, and 150 mg/kg administered twice daily for 7 consecutive days (BID), with a maximum inhibition rate of 75%[1].
