ACTH (1-17) TFA is a potent agonist of hMC1R. ACTH (1-17) has a high affinity for hMC1R with a Ki value of 0.21 ± 0.03 nM, slightly higher than the Ki value of α-MSH (0.13 ± 0.005 nM) [1]. Even when using micromolar concentrations of the peptide in rat pituitary cultures, ACTH (1-17) induces only a slight and insignificant increase in growth hormone secretion [2].

DNA marker inhibition was observed when ACTH (1-17) was injected 2 hours after the start of the circadian rhythm when nocturnal animals become active. When injected during this circadian rhythm, especially at larger doses, DNA marker inhibition lasted for 24 hours. If the same dose of ACTH was injected 4 hours earlier, the duration of inhibition was much shorter[3].
