| Targets |
pNP-G7 targets α-amylase, an enzyme that hydrolyzes α-1,4-glycosidic bonds in starch and glycogen. As a substrate, pNP-G7 is specifically cleaved by α-amylase to produce shorter oligosaccharides and p-nitrophenol. The release of p-nitrophenol can be measured at 405 nm, allowing for the quantification of α-amylase activity.
|
|---|---|
| ln Vitro |
pNP-G7 is used as a substrate for α-amylase in in vitro biochemical assays. It is hydrolyzed by serum α-amylase into shorter 4-nitrophenylated oligosaccharides. The release of p-nitrophenol is measured spectrophotometrically to quantify α-amylase activity. pNP-G7 is widely used in enzymology and clinical diagnostics for measuring α-amylase levels.
|
| ln Vivo |
In vivo data for pNP-G7 is not applicable as it is a reagent used for in vitro biochemical assays rather than a therapeutic compound. pNP-G7 is used in clinical diagnostics to measure α-amylase levels in serum samples. It is not administered to animals for in vivo efficacy studies.
|
| Enzyme Assay |
The in vitro α-amylase assay using pNP-G7 involves incubating the substrate with a sample containing α-amylase. The enzyme hydrolyzes pNP-G7 to release p-nitrophenol, which is measured spectrophotometrically at 405 nm. The rate of p-nitrophenol release is directly proportional to α-amylase activity. The assay is performed in a 96-well plate format with appropriate buffers and incubation conditions.
|
| Cell Assay |
The α-amylase activity assay is performed in a 96-well plate format. Serum or other samples containing α-amylase are incubated with pNP-G7 substrate at 37°C. The reaction is stopped after a defined period, and the absorbance is measured at 405 nm. The α-amylase activity is calculated from a standard curve. The assay is used in clinical diagnostics to measure serum α-amylase levels.
|
| Animal Protocol |
In vivo studies are not applicable for pNP-G7 as it is a reagent used for in vitro assays. The compound is used in clinical diagnostics to measure α-amylase levels in serum samples. It is not administered to animals for in vivo efficacy studies.
|

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2.5 mg/mL (1.92 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2.5 mg/mL (1.92 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
