| Targets |
NSC 15364 targets the voltage-dependent anion channel 1 (VDAC1), a mitochondrial membrane protein. VDAC1 oligomerization is a key step in the mitochondrial pathway of apoptosis, leading to the release of cytochrome c and other apoptotic factors. By preventing VDAC1 oligomerization, NSC 15364 inhibits this apoptotic pathway.

| ln Vitro |
One of the main targets of medications meant to control cells is the oligomerization of mitochondrial membrane protein potential channel 1 (VDAC1) [1].
In vitro, NSC 15364 is an inhibitor of VDAC1 oligomerization and apoptosis. It can be used for screening of ALR inhibitors by detection of hydrogen peroxide production. Its mechanism of action is confirmed by its ability to prevent the formation of VDAC1 oligomers in cellular or cell-free systems. |
| ln Vivo |
In vivo, NSC 15364's ability to inhibit apoptosis makes it a valuable tool for studying diseases characterized by excessive apoptosis. By preventing VDAC1 oligomerization, it could potentially protect cells from apoptotic cell death. Its in vivo efficacy would need to be confirmed in appropriate animal models.
|
| Enzyme Assay |
Non-cellular assays for NSC 15364 involve measuring its ability to prevent VDAC1 oligomerization. This can be done using purified VDAC1 protein and cross-linking agents, followed by SDS-PAGE and Western blot to detect oligomer formation. The compound's direct binding to VDAC1 can be assessed using surface plasmon resonance (SPR) or isothermal titration calorimetry (ITC).
|
| Cell Assay |
In vitro cellular assays for NSC 15364 are conducted to assess its effects on apoptosis. Cells are treated with an apoptotic stimulus (e.g., staurosporine, etoposide) in the presence or absence of NSC 15364. Apoptosis is measured by assessing caspase activation, cytochrome c release, and DNA fragmentation. VDAC1 oligomerization is measured by cross-linking and Western blot.
|
| Animal Protocol |
In vivo animal experiments with NSC 15364 are conducted in models of diseases where apoptosis plays a key role, such as ischemia-reperfusion injury, neurodegeneration, or certain cancers. The compound is administered, and its effects on tissue damage, cell death, and functional recovery are assessed.
|
| ADME/Pharmacokinetics |
Pharmacokinetic data for NSC 15364 are limited. It has a molecular weight of 242.28. It is soluble in DMSO and is typically stored as a powder at -20°C. Its half-life and bioavailability would need to be characterized for in vivo use.
|
| Toxicity/Toxicokinetics |
NSC 15364 is considered to have low toxicity based on its use as a research compound. However, comprehensive toxicological data are not extensively published. The compound is intended for research use only and is not approved for human therapeutic use.

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2.5 mg/mL (10.32 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2.08 mg/mL (8.59 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.8 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
