Naringenin chalcone (CAS# 73692-50-9), also known as Isosalipurpol, is a flavonoid compound and an intermediate in flavonol biosynthesis that is spontaneously metabolized into naringenin by chalcone isomerase. It exhibits anti-allergic, anti-inflammatory, and anti-asthmatic properties. The compound improves adipocyte metabolic functions and exerts insulin-sensitizing effects by activating an adiponectin-related pathway.
Biological Activity I Assay Protocols (From Reference)
Targets
Naringenin chalcone targets multiple pathways. It inhibits the production of proinflammatory cytokines such as TNF-α and MCP-1. It suppresses asthmatic symptoms by inhibiting Th2 cytokine production from CD4 T cells. The compound also inhibits human placental CYP19A1 activity with an IC₅₀ of 2.6 μmol/L. Additionally, it has been shown to induce apoptosis.
ln Vitro
In vitro, naringenin chalcone exhibits anticancer activity against glioblastoma. It inhibits the production of MCP-1 and NO. The compound has anti-inflammatory and anti-allergic properties, and demonstrates an IC₅₀ of 68 μg/mL in certain assays. It improves adipocyte functions by enhancing adiponectin production and has been shown to inhibit CYP19A1 activity with an IC₅₀ of 2.6 μmol/L.
ln Vivo
In vivo, naringenin chalcone has demonstrated anti-inflammatory and antiallergic activity in mouse models. It suppresses asthmatic symptoms by inhibiting Th2 cytokine production. The compound can be taken orally and has shown effectiveness in various pharmacological models. Its insulin-sensitizing effects have been observed through activation of adiponectin-related pathways.
Enzyme Assay
For in vitro enzyme/receptor binding assays, naringenin chalcone can be evaluated for CYP19A1 inhibitory activity. Microsomal preparations containing CYP19A1 are incubated with the substrate and varying concentrations of the compound. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.8 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL.
Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.
