Targets

The compound inhibits inflammatory signaling proteins such as AKT, p38 MAPK, PI3K, NLRP3, FOXO1, and TLR4.
ln Vitro
In vitro, kushenol I increases IL-10 expression and inhibits key inflammatory signaling proteins such as AKT, p38 MAPK, PI3K, NLRP3, FOXO1, and TLR4. It exhibits inhibitory activity against SGLT2 and is active against the plant pathogenic fungus Cladosporium cucumerinum. The compound also exhibits quantifiable estrogenic, anti-inflammatory, and antifungal activities.
ln Vivo
In vivo, kushenol I has demonstrated anti-inflammatory effects in DSS-induced ulcerative colitis mouse models. Oral administration improves colon length, body weight, and disease scores while reducing inflammatory cytokines (IL-1β, IL-6, IL-17, TNF-α) and increasing IL-10 expression. It enhances intestinal barrier integrity, modulates gut microbiota, and elevates splenic T-cell proportions.
Enzyme Assay
For in vitro enzyme/receptor binding assays, kushenol I can be evaluated against multiple targets including GABAA receptors, cytochrome P450 isoforms, and SGLT2. Receptor binding assays using radiolabeled ligands are performed to determine affinity. Enzyme inhibition assays using recombinant enzymes or microsomal preparations are performed with specific substrates, and activity is measured by LC-MS/MS or fluorescence detection.

Solubility in Formulation 1: ≥ 2.5 mg/mL (5.50 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL.
Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.
