| Targets |
Demethoxycurcumin modulates multiple signaling pathways, including Wnt/β-catenin, AMPK, and MAPK. It attenuates the Wnt/β-catenin pathway through down-regulation of the transcriptional coactivator p300. Demethoxycurcumin exerts its anti-inflammatory effect in LPS-activated cells, which may be partly due to its potent down-regulation of NADPH-derived reactive oxygen species (ROS) production.

| ln Vitro |
In vitro, demethoxycurcumin induces the apoptosis of human lung cancer NCI-H460 cells through the mitochondrial-dependent pathway. It regulates anti-inflammatory and anti-proliferative responses through a ROS-independent mechanism. Demethoxycurcumin also has anti-acanthamoebic effects and shows antioxidant activity.
|
| ln Vivo |
In vivo, demethoxycurcumin has demonstrated anticancer, antiproliferative, and antioxidant activities. It may be used as a novel anticancer agent for the treatment of lung cancer in the future. Its anti-inflammatory activity has been studied in various animal models of inflammation.
|
| Enzyme Assay |
In vitro enzyme/receptor binding assays for demethoxycurcumin typically measure its effects on signaling pathways. The compound's ability to inhibit Wnt/β-catenin signaling can be assessed using reporter gene assays. Its effects on AMPK and MAPK pathways can be evaluated by measuring the phosphorylation status of key proteins in these pathways.
|
| Cell Assay |
Cell-based assays for demethoxycurcumin involve treating various cancer cell lines with the compound and measuring cell proliferation, apoptosis, and signaling pathway activation. The compound's ability to induce apoptosis through the mitochondrial-dependent pathway is assessed by measuring caspase activation and mitochondrial membrane potential.
|
| Animal Protocol |
Demethoxycurcumin has been studied in animal models of cancer and inflammation. In xenograft models, the compound's ability to inhibit tumor growth is assessed. Its anti-inflammatory effects are evaluated in models of acute and chronic inflammation.

Metabolism / Metabolites
The known metabolites of demethoxycurcumin include demethoxycurcumin O-glucuronide. Dedicated pharmacokinetic data for demethoxycurcumin is limited. As a curcuminoid, it is expected to have poor oral bioavailability due to rapid metabolism and elimination. The compound is typically dissolved in DMSO for in vitro experiments. |
| Toxicity/Toxicokinetics |
Demethoxycurcumin is generally considered to have low toxicity. It is a natural compound with a favorable safety profile. However, as with any research compound, appropriate safety precautions should be taken when handling it.

| Additional Infomation |
Demethoxycurcumin is a β-diketone, a compound in which one methoxy group of curcumin is replaced by a hydrogen atom. It is found in Curcuma zedoaria and Etlingera elatior. It possesses metabolite, antitumor, and anti-inflammatory activities. It is a polyphenol, β-diketone, enone, and diarylheptane compound. Demethoxycurcumin has also been reported to exist in Curcuma xanthorrhiza, Curcuma kwangsiensis, and other organisms with relevant data.
Demethoxycurcumin (DMC) is one of the main active curcuminoids found in turmeric. It has anti-inflammatory, antioxidant, antibacterial, anticancer, and neuroprotective effects. It modulates multiple signaling pathways, including Wnt/β-catenin, AMPK, and MAPK. It is a valuable research tool for studying these pathways and their roles in cancer and inflammation. |

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 3.25 mg/mL (9.61 mM) (saturation unknown) in 10% DMSO + 40% PEG300 +5% Tween-80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 32.5 mg/mL clear DMSO stock solution to 400 μL of PEG300 and mix evenly; then add 50 μL of Tween-80 + to the above solution and mix evenly; then add 450 μL of normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
