Product Description
Z-DL-Pro-OH (N-Cbz-DL-proline) is a proline analogue that may be utilized in the preparation /synthesis of active molecules or other compounds.
Z-DL-Pro-OH, also known as N-Benzyloxycarbonyl-DL-proline or N-Cbz-DL-proline, is a racemic mixture of protected proline enantiomers. It features a benzyloxycarbonyl (Z or Cbz) group protecting the N-terminus of DL-proline. It is used as a building block in peptide synthesis. The racemic nature means it contains both D- and L-proline, which can be used to study the effects of stereochemistry or can be resolved into its individual enantiomers.
Biological Activity I Assay Protocols (From Reference)
Targets
Z-DL-Pro-OH is classified as a protected amino acid derivative and a proline analogue. It does not have a specific biological receptor target. Its primary application is in peptide synthesis for introducing proline residues. The Z group provides orthogonal N-terminal protection removable by hydrogenolysis. The cyclic structure of proline imparts conformational constraints to peptides.
ln Vitro
In vitro activity for Z-DL-Pro-OH is primarily as a peptide synthesis reagent. Amino acid derivatives have been studied as ergogenic supplements. Z-DL-Pro-OH itself is not directly biologically active. Its utility is in preparing proline-containing peptides for subsequent biological evaluation. The proline residue can induce turns and influence peptide conformation.
ln Vivo
In vivo activity data for Z-DL-Pro-OH as a standalone compound is not applicable. It is a protected amino acid building block used in research. Peptides synthesized using this building block may be evaluated in animal models. The Z protecting group is typically removed before any biological testing.
Enzyme Assay
In vitro protocols for Z-DL-Pro-OH involve its use in peptide synthesis. The compound is activated using coupling reagents and coupled to a growing peptide chain. The Z group is removed by catalytic hydrogenolysis. Purity is typically high. Coupling efficiency is monitored by standard methods.
Cell Assay
Cell-based protocols are not directly applicable to Z-DL-Pro-OH as it is a synthesis building block. Proline-containing peptides synthesized using this reagent can be tested in cell culture. Peptides are dissolved in DMSO or appropriate buffers and added to cells at concentrations of 0.1-100 µM. Incubation is typically 24-72 hours.
Animal Protocol
Animal studies with Z-DL-Pro-OH are not conducted directly. Peptides containing proline residues synthesized using this building block may be evaluated in vivo. Protocols involve administration of the peptide via appropriate routes in animal models. Pharmacokinetic sampling, efficacy assessment, and toxicological evaluation follow standard preclinical study designs.
ADME/Pharmacokinetics
Pharmacokinetic data for Z-DL-Pro-OH as a standalone compound is not applicable. The compound has a molecular weight of 249.26 g/mol and is a solid at room temperature. Storage is recommended in a cool, dry place. The Z group is removed by hydrogenolysis during peptide synthesis.
Toxicity/Toxicokinetics
Limited toxicological data is available for Z-DL-Pro-OH. The compound is for research use only. Standard safety precautions should be observed. No specific toxicity studies have been reported. It is not intended for human therapeutic use.
