PEGs
PROTAC Linkers; Liposomes; Biotin-binding proteins (avidin/streptavidin). |
|---|---|
| ln Vitro |
One ligand is for an E3 ubiquitin ligase, and the other is for the target protein; these two ligands are joined by a linker to form PROTACs. The intracellular ubiquitin-proteasome system is utilized by PROTACs to specifically destroy target proteins[1].
As a PEGylated phospholipid, DSPE-PEG-Biotin itself has no direct biological activity but is a key functional material for drug delivery. It can be incorporated into liposomes or micelles via self-assembly of the DSPE anchor in lipid bilayers. The long PEG chain reduces immunogenicity and prolongs circulation half-life, while the biotin enables targeting or capture through avidin-biotin interactions. |
| ln Vivo |
The in vivo activity of DSPE-PEG-Biotin is realized when it is formulated as part of a nanocarrier system. The compound enhances the stability and circulation time of liposomes in vivo by generating steric hindrance and electrostatic repulsion. Biotin can target the sodium-dependent multivitamin transporter (SMVT) on tumor cells, facilitating active tumor targeting.
|
| Enzyme Assay |
DSPE-PEG-Biotin is not used in standard enzyme/receptor binding assays. However, its biotin group is routinely used in biophysical assays such as surface plasmon resonance (SPR) or enzyme-linked immunosorbent assays (ELISA) by capturing the biotinylated liposomes onto a streptavidin-coated sensor surface.
|
| Cell Assay |
This linker is not tested directly in cell-based assays. Instead, cells are treated with liposomes or nanoparticles containing this compound. Uptake of biotinylated nanoparticles into cells is often evaluated using confocal microscopy or flow cytometry by labeling them with a fluorescent streptavidin-conjugated probe.
|
| Animal Protocol |
For in vivo studies, DSPE-PEG-Biotin is incorporated into lipid nanoparticles (LNPs). These LNPs are typically administered via intravenous (IV) injection in animal models, often at doses ranging from 0.5 to 5 mg/kg based on lipid content. Tissue distribution and pharmacokinetics are measured by extracting lipids from tissues and quantifying them by LC-MS or using radiolabeled DSPE.

| Additional Infomation |
DSPE-PEG-Biotin is widely used for constructing biosensors, affinity purification systems, and targeted lipid vesicles. The long PEG spacer (MW 2000) improves biotin accessibility while maintaining bilayer integrity, ensuring consistent performance in surface chemistry and biointerfacial applications. It is also classified as a PROTAC linker of the PEG category.

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2.5 mg/mL (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween-80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), Clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of DMSO stock solution (25.0 mg/mL) to 400 μL of PEG300 and mix well; then add 50 μL of Tween-80 and mix well; finally add 450 μL of physiological saline and adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2.5 mg/mL (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), Clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of DMSO stock solution (25.0 mg/mL) to 900 μL of 20% SBE-β-CD saline and mix well. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution. Solubility in Formulation 3: ≥ 2.5 mg/mL (saturation unknown) in 10% DMSO + 90% Corn Oil (add these co-solvents sequentially from left to right, and one by one), Clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of DMSO stock solution (25.0 mg/mL) to 900 μL of corn oil and mix well.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
