| Enzyme Assay |

Terminate by adding 0.5% TFA. Analyze by MALDI-TOF or LC-MS to detect mass increase due to methylation. For ELISA-based detection, coat the peptide on a plate and use anti-H3K27me3 antibody. For a binding assay, immobilize the peptide on a streptavidin plate (if biotinylated) and add a reader protein (e.g., CBX7), detect with specific antibody. The TFA salt is soluble; dissolve in water.
