Targets
OVA (241-270) TFA contains multiple T cell epitopes that can be recognized by both CD4+ and CD8+ T cells after processing. In the context of antigen-presenting cells, this peptide is taken up, processed, and presented on MHC class I and class II molecules. It activates OVA-specific T cells (particularly OT-I and OT-II T cells) in a non-specific manner. This peptide enables the study of T cell activation, proliferation, and cytokine production. It is a non-specific activator of cytotoxic T lymphocytes (CTLs). It is a fragmented peptide of the OVA antigen. This peptide plays an important role in the immune system by activating CTLs, which are specialized immune cells that identify and kill infected or abnormal cells.
ln Vitro
In vitro, OVA (241-270) TFA is processed and presented to T cells after uptake by dendritic cells (DCs) or other antigen-presenting cells (APCs). Unlike the minimal epitope peptide OVA (257-264) which directly loads onto MHC class I, the longer OVA (241-270) peptide requires intracellular processing. In cell-based assays, bone marrow-derived dendritic cells (BMDCs) are pulsed with OVA (241-270) (10-100 microg/mL) for 2-4 hours, washed, and cocultured with OVA-specific CD8+ T cells (OT-I) or CD4+ T cells (OT-II). T cell activation is measured by CD69 expression, proliferation (CFSE dilution), or cytokine production (IFN-gamma, IL-2). The peptide can elicit responses from both CD4+ and CD8+ T cells due to the presence of overlapping epitopes.
ln Vivo
In vivo, OVA (241-270) TFA can be used to prime T cell responses in C57BL/6 mice. Mice are immunized subcutaneously or intraperitoneally with OVA (241-270) peptide (50-200 microg per mouse) emulsified in Complete Freund's Adjuvant (CFA) or mixed with other adjuvants. Alternatively, the peptide can be delivered in liposomes or nanoparticles. After 7-14 days, splenocytes or lymph node cells are harvested and restimulated ex vivo with the peptide. T cell responses are analyzed by intracellular cytokine staining, ELISpot, or cytotoxicity assays. The longer peptide may be processed differently than the minimal epitope, providing insights into antigen processing pathways. The peptide also contributes to the induction of delayed-type hypersensitivity (DTH) in BALB/c mice.
Enzyme Assay
OVA (241-270) TFA is not used in cell-free enzyme assays. For receptor binding assays, it is not directly used because the peptide requires processing. However, a T cell hybridoma activation assay can be performed to study antigen presentation. BMDCs or spleen-derived APCs are seeded in 96-well plates (1×10⁵ cells/well). OVA (241-270) TFA (0.1-100 microM) is added and incubated for 2-4 hours. After washing, OVA-specific T cell hybridomas (e.g., B3Z for CD8+ response or other hybridomas for CD4+ response) are added at 1×10⁵ cells/well. After 18-24 hours, activation is measured by IL-2 production (ELISA) or lacZ expression (using a colorimetric substrate for beta-galactosidase, e.g., CPRG). The EC50 for T cell activation is calculated from the dose-response curve.
Cell Assay
For T cell proliferation assays, CD8+ T cells are isolated from OT-I transgenic mice (or CD4+ T cells from OT-II mice). Splenocytes are enriched using magnetic bead-based negative selection. Isolated T cells are labeled with CFSE (5 microM, 10 min, 37degC). APCs (irradiated splenocytes or BMDCs) are incubated with OVA (241-270) TFA (1-100 microg/mL) for 2 hours, then washed extensively to remove free peptide. CFSE-labeled T cells are added to APCs at a 1:1 ratio. After 72 hours, T cell proliferation is measured by CFSE dilution using flow cytometry. Alternatively, T cell activation is assessed by CD69 and CD25 expression at 24 hours. For cytokine production, culture supernatants are collected at 48 hours and analyzed by ELISA for IFN-gamma, IL-2, TNF-alpha (for CD8+), and IL-4, IL-5, IL-13 (for CD4+ Th2 responses). Female BALB/c mice are used for DTH studies. OVA (241-270) TFA is involved in the generation and development of delayed-type hypersensitivity reactions.
Animal Protocol
For immunization studies, female C57BL/6 mice (6-8 weeks old, n=5-8 per group) are immunized subcutaneously at the base of the tail or in the flank with OVA (241-270) TFA (100 microg per mouse) emulsified in CFA (1:1 volume). A control group receives CFA alone. On day 7, mice receive a booster immunization with the same dose of peptide in Incomplete Freund‘s Adjuvant (IFA). On day 14, spleens and draining lymph nodes (inguinal and popliteal) are harvested. Single-cell suspensions are prepared. For antigen-specific recall responses, 2×10⁵ splenocytes are cultured with OVA (241-270) peptide (0-100 microg/mL) for 48-72 hours. Supernatants are collected for cytokine measurement by multiplex assay or ELISA. Alternatively, an ELISpot assay is performed to enumerate IFN-gamma or IL-4 spot-forming cells. For DTH studies, BALB/c mice are primed with OVA (241-270) (100 microg in CFA, sc). On day 7, mice are challenged intradermally in the footpad or ear with the peptide (50 microg in 50 microL PBS). DTH response is measured as footpad swelling (using a caliper) or ear thickness at 24 hours. This peptide is important for DTH responses.
ADME/Pharmacokinetics
The TFA salt has a molecular weight of 3535.92 Da with formula C156H247F3N34O53S1. The lyophilized powder should be stored at -80degC (2 years) or -20degC (1 year) in sealed containers away from moisture. For solution storage, it should be kept at -80degC for 6 months or -20degC for 1 month under sealed, moisture-protected conditions. The peptide is typically soluble in DMSO (10-20 mg/mL) and PBS. The TFA salt enhances solubility. Working solutions should be prepared fresh or stored in small aliquots to avoid repeated freeze-thaw cycles. Avoid prolonged exposure to light and moisture.
