In female mice of the H‑2ᵇ haplotype (e.g., C57BL/6), a single subcutaneous or intraperitoneal injection of Uty HY peptide (50‑200 ug) emulsified in complete Freund's adjuvant (CFA) induces a strong CD8+ T‑cell response against male cells. When these immunized female mice are subsequently transplanted with male skin grafts, the grafts are rejected in an accelerated, T‑cell‑dependent manner (median survival time <20 days compared to >50 days in non‑immunized females).

| Enzyme Assay |
A direct binding assay between the peptide and H‑2Dᵇ molecules is performed using surface plasmon resonance (SPR) or a competition‑based refolding assay. For the refolding assay, recombinant H‑2Dᵇ heavy chain and beta2‑microglobulin are denatured and then diluted into refolding buffer containing the Uty HY peptide (10‑100 uM). The refolded MHC‑peptide complex is detected by a conformation‑specific antibody (e.g., 28‑14‑8S). Alternatively, RMA‑S cells (a transporter‑associated with antigen processing‑deficient cell line) can be pulsed with the peptide and tested for surface H‑2Dᵇ‑peptide complexes by flow cytometry.

| Cell Assay |
RMA‑S cells (H‑2ᵇ) are incubated with the Uty HY peptide (0.01‑100 uM) overnight at 26degC to allow exogenous peptide binding to empty MHC I molecules. The cells are then shifted to 37degC for 2 hours to stabilize the complexes. Surface expression of H‑2Dᵇ‑peptide complexes is detected by staining with an anti‑H‑2Dᵇ antibody (e.g., clone 34‑2‑12) and analyzed by flow cytometry. For T‑cell activation assays, female splenocytes are cultured with peptide‑pulsed and irradiated male splenocytes for 5 days, then the cytotoxic activity is tested.

C57BL/6 female mice (6‑8 weeks old) are immunized subcutaneously at the base of the tail with 100 ug of Uty HY peptide emulsified in 100 uL of CFA. After 7‑10 days, the mice are challenged by subcutaneous transplantation of a full‑thickness skin graft from a male C57BL/6 donor. Graft survival is monitored daily by visual inspection and palpation; rejection is defined as >90% graft necrosis. Alternatively, splenocytes from immunized mice can be harvested and tested for ex vivo cytotoxicity.

| ADME/Pharmacokinetics |
The peptide itself has a very short half‑life in plasma (<10 minutes) due to rapid proteolysis. The TFA salt does not change this. The 9‑mer peptide is small enough to be filtered by the kidney and degraded in the renal tubules. To induce T‑cell responses, the peptide must be formulated in an adjuvant (e.g., CFA) that prolongs its presence at the injection site and activates innate immunity.

| Toxicity/Toxicokinetics |
The Uty HY peptide is non‑toxic at the doses used in mice (<200 ug). The CFA emulsion can cause local injection site reactions (granulomas) and, rarely, systemic inflammatory responses. The TFA counterion does not contribute to toxicity at these concentrations. No genotoxicity or organ toxicity has been reported.
