Human Endogenous Metabolite
D-Arginine targets arginine decarboxylase (EC 4.1.1.19) as an inhibitor. It is the D-isomer of arginine and is considered an inactive form of L-arginine. It is an endogenous metabolite. It has broad-spectrum toxicity to microorganisms and acts as a regulator of microbial subpopulations.
ln Vitro
In vitro, D-Arginine is the D-isomer and inactive form of L-Arginine. It inhibits arginine decarboxylase (EC 4.1.1.19). It has broad-spectrum toxicity to microorganisms and acts as a regulator of microbial subpopulations. It is used as an organic substrate to study the metabolic versatility of freshwater sedimentary archaea.
ln Vivo
In vivo, D-Arginine is a semi-essential amino acid that maintains cell death in the central nervous system (CNS). It may possess neuroprotective actions against glucocorticoids induced neurotoxicity in the CNS. It inhibits the antinociceptive activity of L-arginine in a mouse model of carrageenan-induced pain when administered at a dose of 1,000 mg/kg.
Enzyme Assay
In vitro enzyme assays for arginine decarboxylase involve incubating the enzyme with D-Arginine as inhibitor and L-arginine as substrate in buffer at pH 7.4. The production of agmatine is quantified by HPLC or LC-MS/MS. Inhibition constants are determined from dose-response curves. The compound's effects on microbial growth are assessed by measuring optical density in bacterial cultures.
Cell Assay
For in vitro cell assays, cells are cultured in medium supplemented with D-Arginine at concentrations of 0.1-10 mM. Cell viability is assessed by MTT assay. The compound's effects on arginine metabolism and nitric oxide production are evaluated by measuring arginine and citrulline levels. Neuroprotective effects are assessed in neuronal cell models treated with glucocorticoids.
Animal Protocol
For in vivo animal studies, D-Arginine is administered to rodents via oral gavage or intraperitoneal injection at doses of 10-1000 mg/kg.
