NMN itself acts as a metabolite with biological activities on endothelial cells.
ln Vitro
N-Methylnicotinamide exhibits antithrombotic effects via production and release of prostacyclin, which inhibits arterial thrombosis development. It has multiple effects on endothelial cells: it inhibits thrombus formation, increases skin vascular permeability, and prevents atherosclerosis in rodent models. NMN has been shown to protect against endothelial dysfunction and attenuate atherogenesis in animal models.
ln Vivo
In vivo, N-Methylnicotinamide has been shown to prevent atherosclerosis in rodent models. It protects against endothelial dysfunction and attenuates atherogenesis. Enzyme Assay
In vitro enzyme assays for NNMT involve incubating the enzyme with N-methylnicotinamide and S-adenosylmethionine (SAM) as methyl donor in Tris-HCl buffer at pH 7.4.
