| Targets |
Mitochondrial hypochlorous acid (HOCl) is the primary molecular target. The probe is designed to selectively target mitochondria and detect HOCl within this organelle.
|
|---|---|
| ln Vitro |
HKOCl-4m (5 μM; 30 min): When HOCl was added, the fluorescence intensity dramatically increased. It has been effectively possible to identify endogenous HOCl in RAW264.7 murine macrophages by using HKOCl-4m. In live cells, the production of mitochondrial HOCl can be seen using HKOCl-4m [1].
HKOCl-4m (5 uM; 30 min) exhibits a robust increase in fluorescence intensity upon addition of HOCl in cell-free assays. It is a rhodol-based fluorescent probe that selectively targets mitochondria for monitoring mitochondrial HOCl. Its maximum absorption/emission wavelength is 490/527 nm. |
| ln Vivo |
There is no specific in vivo activity data available for this compound. However, as a fluorescent probe for reactive species, similar compounds are typically used for real-time imaging of HOCl in live cells and animal models.
|
| Enzyme Assay |
A standard protocol involves incubating HKOCl-4m with varying concentrations of HOCl in a buffer solution (e.g., PBS, pH 7.4) for a short period at 37degC. The fluorescence intensity ratio at specific wavelengths is then measured using a spectrophotometer or microplate reader.
|
| Cell Assay |
A general protocol involves seeding cells in a confocal dish and incubating with HKOCl-4m (e.g., 5 uM) for 30 minutes at 37degC. The mitochondrial co-localization can be confirmed using a commercial mitochondrial tracker. After washing, cells are treated with an HOCl donor, and ratiometric fluorescence imaging is performed.
|
| Animal Protocol |
A typical protocol for in vivo imaging involves intravenous or intraperitoneal injection of an optimized concentration of HKOCl-4m into a live animal model. After a suitable circulation time (e.g., 30 minutes to 2 hours), the animal is placed under an in vivo imaging system to monitor HOCl-induced fluorescence.
|
| ADME/Pharmacokinetics |
General PK properties for small molecule probes are not applicable, as these compounds are typically used for acute imaging. However, formulation can be in PBS or saline with co-solvents like DMSO for in vivo use.
|
| Toxicity/Toxicokinetics |
General toxicity for imaging probes is considered low. For HKOCl-4m, no specific toxicity studies are reported. In similar experiments, incubation with up to 10 uM for 24 hours in cell models shows good biocompatibility and no significant cytotoxicity.

| Additional Infomation |
HKOCl-4m is a research tool for studying oxidative stress and inflammatory diseases, particularly those involving mitochondrial reactive species. The ratiometric detection offers built-in correction for probe concentration and environmental factors.

| Solubility (In Vivo) |
Solubility in Formulation 1: 2.63 mg/mL (2.28 mM) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), suspension solution; with sonication.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 26.3 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
