| Targets |
The primary targets of methyltetrazine-amine hydrochloride are not biological targets but rather chemical reaction partners in bioorthogonal labeling applications. The methyltetrazine group specifically reacts with strained alkynes (e.g., trans-cyclooctene, bicyclononyne) via IEDDA cycloaddition. This reaction is highly selective and rapid, making it suitable for labeling biomolecules in complex biological environments. The amine group provides a handle for conjugation to proteins, antibodies, or other biomolecules via amide bond formation.

| ln Vitro |
In vitro activity of methyltetrazine-amine hydrochloride refers to its chemical reactivity in bioorthogonal labeling reactions. The compound demonstrates rapid and selective reaction with strained alkynes via IEDDA cycloaddition. The reaction kinetics are among the fastest for bioorthogonal reactions, with second-order rate constants typically in the range of 10-100 M-1s-1. The compound can be used to label biomolecules that have been functionalized with strained alkynes, enabling detection or purification of the labeled species.
|
| ln Vivo |
In vivo activity data for methyltetrazine-amine hydrochloride are related to its use in bioorthogonal labeling in living systems. The compound can be used for pretargeted imaging or therapy applications where a tetrazine-modified biomolecule is administered followed by a radiolabeled or drug-loaded trans-cyclooctene probe. The rapid and selective reaction enables specific labeling of target tissues or cells in vivo. These applications have been demonstrated in preclinical imaging and therapeutic studies.

| Enzyme Assay |
Non-cellular assays for methyltetrazine-amine hydrochloride typically involve characterizing its reactivity in IEDDA cycloaddition reactions. The reaction kinetics with various strained alkynes are measured using UV-Vis spectroscopy or HPLC. The compound's stability in biological media is assessed by incubating it in serum or buffer and monitoring degradation over time. These cell-free systems allow for precise characterization of the compound's chemical properties and reactivity.
|
| Cell Assay |
Cellular assays for methyltetrazine-amine hydrochloride are conducted in cell culture to evaluate its bioorthogonal labeling efficiency. Cells expressing or modified with strained alkyne-functionalized biomolecules are treated with the tetrazine compound, and labeling efficiency is assessed by fluorescence microscopy or flow cytometry if the compound is fluorescently labeled. Cytotoxicity of the compound is evaluated using standard cell viability assays to ensure compatibility with live-cell experiments.
|
| Animal Protocol |
In vivo animal experiments for methyltetrazine-amine hydrochloride typically involve pretargeted imaging or therapy studies. The compound (or a conjugate) is administered to mice bearing tumors or other targets that express strained alkyne-functionalized biomolecules. This is followed by administration of a radiolabeled or drug-loaded trans-cyclooctene probe. Imaging or biodistribution studies are performed to assess target-specific accumulation. Toxicity and pharmacokinetics are also evaluated in these studies.
|
| ADME/Pharmacokinetics |
Pharmacokinetic properties of methyltetrazine-amine hydrochloride depend on its formulation and administration route. As a small molecule, it may be rapidly cleared from circulation. For in vivo pretargeting applications, the compound is typically administered intravenously, and its clearance kinetics are optimized to achieve a favorable target-to-background ratio. The compound's stability in blood and its distribution to tissues are important factors for successful in vivo labeling.
|
| Toxicity/Toxicokinetics |
Dosing and toxicity studies of methyltetrazine-amine hydrochloride in animal models have shown that the compound is generally well-tolerated at the doses used for imaging applications. The compound is not intended for therapeutic use at high doses. Standard laboratory safety precautions should be followed when handling the compound. Its safety profile is favorable for research applications, with no significant toxicity reported at typical labeling concentrations.

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2.5 mg/mL (10.52 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2.5 mg/mL (10.52 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |
