Natural product; insect sex pheromone
The Bombykol receptor, a specific odorant receptor (OR) expressed in the antennae of male silkworm moths. Bombykol is a sex pheromone that binds to its specific receptor on the olfactory neurons of male moths. This binding triggers a signal transduction cascade that leads to the attraction of the male towards the female. The interaction between Bombykol and its receptor is a model system for understanding the molecular basis of pheromone perception and insect behavior. |

| ln Vitro |
This study aimed to explore the interaction between bombykol and BmOR1 and also provide a paradigm for agroforestry pest control. The electrochemical biosensor signal amplification system was used: nanogold with horseradish peroxidase. An electrochemical bilayer nanogold membrane receptor sensor was developed using the following schemes and processes: twice self-assembly of nanogold and succeeding absorption of Bombyx mori olfactory receptor 1 (BmOR1); sex pheromone-binding protein; spectral scanning and transmission electron microscope to characterize nanogold sol; and atomic force microscope, cyclic voltammetry, and AC impedance methods to characterize individual processes of sensor assembly. The amperometric I-T curve was adopted to measure the response current upon interaction with different concentrations of bombykol (diluted in phosphate-buffered saline) and BmOR1. The results demonstrated the receptor-ligand interaction pattern, which was similar to enzymatic reaction kinetics, with the activation constant Ka of up to 8.57 × 10-20 mol/L and signal magnification of about 10,000-fold. In this study, the simulation of intracellular receptor signaling cascade by an electrochemical signal amplification system helped in directly measuring BmOR1-bombykol ligand interaction and exploring the kinetics after the self-assembly of BmOR1 on the biosensor. It provided a novel platform for future studies on receptor-ligand interaction [1].
In vitro, Bombykol is used in biological studies to investigate the specificity of pheromone-binding proteins and its interaction with its receptor. Its binding affinity to pheromone-binding proteins (PBPs) and the odorant receptor can be measured using techniques like fluorescence binding assays or electrophysiology (e.g., electroantennogram, EAG). |
| ln Vivo |
In vivo, Bombykol is a behaviorally active compound. When released by the female, it elicits a strong attraction response in male moths in a wind tunnel or in the field. This behavioral response is the primary "activity" measured for this pheromone.
|
| Enzyme Assay |
The activity of Bombykol is assessed using behavioral assays, such as the wind tunnel assay or field trapping experiments. In a wind tunnel, male moths are released, and their ability to fly upwind towards a source of Bombykol is observed and quantified. Electroantennogram (EAG) recordings can measure the electrical response of the antenna to Bombykol.
|
| Cell Assay |
Bombykol is not typically used in cell culture for pharmacological studies. However, insect cell lines expressing the Bombykol receptor can be used to study receptor activation and signaling.
|
| Animal Protocol |
Bombykol is used in behavioral studies with silkworm moths. The compound is applied to a lure or dispenser, and the attraction of male moths is observed in a laboratory or field setting.
|
| ADME/Pharmacokinetics |
Bombykol has a molecular weight of 238.41 g/mol and a molecular formula of C16H30O. It is a colorless liquid or low-melting solid. It is soluble in organic solvents such as ethanol and hexane but practically insoluble in water. It should be stored at low temperatures, protected from light and air, to prevent oxidation.

| Toxicity/Toxicokinetics |
Intraperitoneal LD50 in mice: 6500 mg/kg. Chemotherapy, 13(161), 1968 [PMID:5660663]
Bombykol is considered to have low toxicity to mammals and is not classified as a hazardous substance in the context of its use as a research chemical. However, standard laboratory safety precautions should be followed when handling the compound. |

| Additional Infomation |
Bombykol is a long-chain primary fatty alcohol that functions as a pheromone. It has been reported that Bombykol is found in both the domestic silkworm (Bombyx mori) and the European red pine (Hemileuca electra), and relevant data are available for reference.
Bombykol is a historic and iconic molecule in the field of chemical ecology. It was the first insect pheromone to be identified, and its discovery paved the way for the identification of pheromones in countless other insect species. It remains a valuable tool for studying insect olfaction, behavior, and the evolution of chemical communication. It is not a drug and has no therapeutic applications. |
