SF-3-030 is a highly potent, selective, and non-ATP-competitive ERK1/2 inhibitor. SF-3-030 selectively induces apoptosis in melanoma cells containing mutated BRaf genes and with sustained activation of the ERK1/2 signaling pathway. SF-3-030 can alleviate various asthma symptoms in a mouse asthma model. SF-3-030 can be used for research in asthma and melanoma.
Biological Activity I Assay Protocols (From Reference)
ln Vitro
SF-3-030 (2.5-60 μM; 30 min) selectively inhibited ERK2-mediated c-Fos F site-dependent phosphorylation of Thr325 residues in vitro, with no effect on D-domain-containing substrates (RSK1) or p38α-mediated ATF2 phosphorylation [2]. SF-3-030 (1-100 μM; 20 min-5 h) inhibited the activity of AP-1 and SRE promoters in EGF/PMA-stimulated HeLa/HEK293 cells and inhibited basal AP-1 activity in A375 melanoma cells, with stronger inhibitory efficacy against PMA-induced responses than against EGF-induced responses [2]. SF-3-030 (5-100 μM; 30 min-1 h) selectively inhibited the expression of EGF/PMA-induced F-site-containing IEGs (c-Fos, Fra1, FosB, FosB2, c-Myc) in HeLa and A375 cells, with minimal effect on F-site-free IEGs (c-Jun, Egr1) [2]. SF-3-030 (1-100 μM; 24 h) selectively inhibited the proliferation of constitutive ERK1/2-active melanoma cells (A375, RPMI7951) and induced their apoptosis, with GI50 values of 7.1 μM and 4.6 μM, respectively, while also showing selectivity for Jurkat cells (GI50 > 50 μM) [2].
ln Vivo
SF-3-030 (10 mg/kg; administered 30 minutes before each house dust mite (HDM) stimulation; 5 days a week for 3 weeks) reduced a variety of house dust mite-induced asthma features in mice induced by house dust mite (HDM) [1].

Experimental Results: In EGF-stimulated HeLa cells, the induction of c-Fos, FosB, FosB2, Fra1, and c-Myc was inhibited in a dose-dependent manner. The effect on c-Jun and Egr1 was minimal, and c-Fos phosphorylation at Ser32 in EGF-stimulated HeLa cells was inhibited, but not at Ser374. It inhibited c-Fos phosphorylation at Thr232 and Ser32, but did not inhibit phosphorylation at Ser374, and reduced the expression of FosB, FosB2, Fra1 and c-Myc in PMA-stimulated HeLa cells.
Western Blot analysis [2]

Experimental Results: Inhibited the basal expression of c-Fos, FosB, FosB2, Fra1 and c-Myc, but did not inhibit Egr1.
Animal Protocol
Animal/Disease Models:Balb/c mice (8-10 weeks old) were stimulated by house dust mite (HDM) [1]
Doses: 10 mg/kg
Route of Administration: Administered within 30 minutes before each HDM stimulation; 5 days a week for 3 weeks
Experimental Results: Significantly reduced HDM-induced airway inflammation. Inhibited airway remodeling (AR) induced by repeated allergen stimulation. Inhibited the development of HDM-induced airway inflammatory hyperresponsiveness (AHR). Regulated lung tissue gene expression profile. Regulated the expression profile of multiple pathway-related proteins in lung tissue.
