Deoxytopsentin (compound 5) (10 μM; 240 nM) effectively inhibited recombinant Staphylococcus aureus PK with an IC50 value of 240 nM, and its selectivity for Staphylococcus aureus PK at a concentration of 10 μM was significantly higher than that for human PK isoenzymes [1]. Deoxytopsentin has in vitro antibacterial activity against both methicillin-sensitive and methicillin-resistant Staphylococcus aureus strains, and its activity against three methicillin-resistant Staphylococcus aureus (MRSA) strains is comparable to or stronger than that of oxacillin [1]. Deoxytopsentin binds to the hydrophobic small interface binding pocket of MRSA PK and forms predicted π-π stacking and hydrogen bond interactions with key amino acid residues in the pocket [1].
