At 15 μM, it can increase net ultrafiltration by up to 15%, with an EC50 of 4.2 μM, and has no effect on solute transport or AQP1 expression [1].

Doses: 0.75 μM; 1.5 μM; 7.5 μM; 15 μM; 30 μM Route of Administration: Intravenous injection; single injection 30 minutes before dialysis test Experimental Results: In wild-type mice, net ultrafiltration increased in a dose-dependent manner, with a maximum increase of 15% at 15 μM, and an estimated EC50 of 4.2 μM. In wild-type mice, the initial ultrafiltration rate increased by about 50% (32.2 μL/min at 15 μM). At a concentration of 15 μM, the intraperitoneal fluid volume increased significantly at 30, 60 and 90 minutes. In wild-type mice, sodium sieving was enhanced, with a greater decrease in dialysate sodium concentration at 30 minutes (11.2 mEq/L) at 15 μM concentration. There was no effect on net or initial ultrafiltration rate in Aqp1 knockout mice. In wild-type mice, there were no alterations to osmotic gradient, small solute (urea and glucose) transport, AQP1 protein/mRNA expression, or AQP1 plasma membrane localization. |
