ADAR1i-124 is an A-to-I RNA editing inhibitor that works by inhibiting the catalytic activity of ADAR1p150 and ADAR1p110. ADAR1i-124 activates the type I interferon (IFN) and ZBP1 pathways and inhibits the viability of various cancer cell lines in a dose-dependent manner. ADAR1i-124 induces apoptosis and necrotizing apoptosis. ADAR1i-124 has potential applications in cancer research, such as in cutaneous melanoma and ovarian cancer.
Biological Activity I Assay Protocols (From Reference)
ln Vitro
ADAR1i-124 (0-200 μM; 2 h) effectively inhibited ADAR1p150-mediated A-to-I RNA editing in vitro, with IC50 values of 8.8 μM (position 2) and 6.5 μM (position 5), respectively [1]. ADAR1i-124 (300 μM; 2 h) did not inhibit ADAR2-mediated A-to-I RNA editing in vitro [1]. ADAR1i-124 selectively binds to ADAR1p110 with a higher affinity than ADAR2 and does not bind to GAPDH [1]. ADAR1i-124 (10 μM; 12 h) induced unedited double-stranded RNA co-localized with MDA5 in the filamentous structure of mouse Yumm1.7 cutaneous melanoma cells [1]. ADAR1i-124 (10 μM; 12 h) reduced A-to-I editing in Yumm1.7 cells and promoted the conversion of certain double-stranded RNAs from type A to type Z [1]. ADAR1i-124 (48 h) showed dose-dependent cytotoxicity against mouse (Yumm1.7 CM, HSG2 OC) and human (WM4223 AM, WM3000 CM) cancer cells, with IC50 values of 2.3, 1.2, 1.1 and 6.6 μM, respectively [1]. ADAR1i-124 (10 μM; 24 h) activated the IFN signaling pathway in Yumm1.7 melanoma cells and HGS2 OC cells [1]. ADAR1i-124 (10 μM; 24 h) can induce apoptosis in HeLa and Yumm1.7 cells, and can also induce necrotic apoptosis in Yumm1.7 cells activated by the ZBP1 pathway [1].

For example, if 1 mL of working solution is to be prepared, you canAdd 100 μL of DMSO stock solution (25.0 mg/mL) to 400 μL of PEG300 and mix well; then add 50 μL of Tween-80 and mix well; finally add 450 μL of physiological saline and adjust the volume to 1 mL.
