In mixed human liver microsomes, the generation of 5-hydroxytryptamine-O-glucuronide (0.025 mg/mL HLM protein; 20-10000 µM 5-hydroxytryptamine; 20 min) catalyzed by UGT1A6 showed weak substrate inhibition kinetics. The average Km value was 420 µM without the addition of 2% BSA and 330 µM after the addition of 2% BSA. The addition of BSA had little effect on the free intrinsic clearance rate [1]. Under the catalysis of recombinant UGT1A6, the generation of 5-hydroxytryptamine-O-glucuronide (20-10000 µM 5-hydroxytryptamine; 90 min) catalyzed by 0.025 mg/mL rUGT1A6 protein also showed weak substrate inhibition kinetics. The average Km value was 570 µM without the addition of 2% BSA and 490 µM after the addition of 2% BSA. The addition of BSA slightly increased the effect on the intrinsic clearance rate [1].
