| Targets |
Neuropeptide Y (3-36) targets the neuropeptide Y Y2 receptor with high selectivity. The Y2 receptor is a G-protein coupled receptor that functions as a presynaptic autoreceptor, inhibiting the release of norepinephrine and other neurotransmitters. The peptide shows minimal activity at Y1 and Y5 receptors.
|
|---|---|
| ln Vitro |
In vitro, Neuropeptide Y (3-36) binds selectively to the Y2 receptor. It reduces the release of norepinephrine via activation of the Y2 receptor. The peptide is used to study NPY receptor pharmacology and the role of Y2 receptors in neurotransmission and metabolic regulation.
|
| ln Vivo |
In vivo, Neuropeptide Y (3-36) decreases the release of norepinephrine via the Y2 receptor. It is involved in the regulation of food intake, energy balance, and cardiovascular function. The peptide has been shown to reduce food intake when administered to animal models, in contrast to the orexigenic effects of full-length NPY.
|
| Enzyme Assay |
Receptor binding assays for Neuropeptide Y (3-36) use membrane preparations from cells expressing Y1, Y2, Y4, or Y5 receptors. Radiolabeled NPY is incubated with varying concentrations of the test peptide, and binding affinity is determined by competition binding curves.
|
| Cell Assay |
Cellular assays for Neuropeptide Y (3-36) typically use cell lines expressing the Y2 receptor. Cells are treated with the peptide, and downstream signaling pathways such as cAMP inhibition or calcium mobilization are measured to confirm receptor activation.
|
| Animal Protocol |
Norepinephrine release assays in brain slices or synaptosomes are used to assess functional activity. In vivo studies are conducted in rodent models, with administration via intracerebroventricular or peripheral injection. Food intake, blood pressure, and neurotransmitter levels are monitored.
|
| ADME/Pharmacokinetics |
As a peptide, Neuropeptide Y (3-36) is expected to have a short half-life due to proteolytic degradation. The compound is a metabolite of full-length NPY, produced by DPP4 cleavage. Standard peptide storage conditions apply.
|
| Toxicity/Toxicokinetics |
Toxicity data for Neuropeptide Y (3-36) are limited. Peptide neuropeptides are generally well-tolerated at physiological concentrations. Standard safety precautions for laboratory handling of research compounds should be followed.

| Additional Infomation |
Neuropeptide Y (3-36) is a research-use peptide and is not approved for therapeutic applications. It is used to study NPY receptor pharmacology, particularly the role of the Y2 receptor in norepinephrine release, food intake regulation, and energy homeostasis. It is available from multiple research suppliers.

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2.5 mg/mL (0.62 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2.5 mg/mL (0.62 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
