| Targets |
TRPV1 (transient receptor potential vanilloid 1) receptor. Liquiritin apioside is an orally active inhibitor of TRPV1 receptor. It inhibits LCR by inhibiting reactive oxygen species (ROS) and NADPH oxidase, weakening the interaction between ROS and TRPV1. TRPV1 is a non-selective cation channel activated by capsaicin, heat, and protons, playing a role in pain and reflex responses.
|
|---|---|
| ln Vitro |
Liquiritin apioside (0.1 mM; overnight) significantly changes the electrophysiological kinetics of laryngeal C neuron currents induced by capsaicin, reducing the current amplitude, increasing the rise time, and prolonging the decay time. It inhibits capsaicin-induced currents in laryngeal C-neurons. The compound also modulates oxidative stress, regulates neurotransmitter balance, and suppresses pro-inflammatory mediators.
|
| ln Vivo |
Liquiritin apioside (10 μg/μL, intralaryngeal or perineural administration; single dose) significantly attenuates the apnea, hypertension, and bradycardia responses to the laryngeal chemoreflex induced by capsaicin, hydrochloric acid, and distilled water in rat pups, but has no significant effect on the laryngeal mechanoreflex. The compound is orally active and has been studied for respiratory-related diseases.
|
| Enzyme Assay |
In vitro receptor binding assays for Liquiritin apioside can be performed using cells expressing recombinant TRPV1 receptors. Radioligand binding assays using [3H]resiniferatoxin or [3H]capsaicin can be used to assess competitive binding. Calcium flux assays using fluorescent calcium indicators (e.g., Fura-2, Fluo-4) are commonly used to measure TRPV1 activation by capsaicin and inhibition by Liquiritin apioside.
|
| Cell Assay |
Cellular assays for Liquiritin apioside are performed using primary cultures of laryngeal C-neurons isolated from nodose/jugular ganglia of rat pups. Cells are treated with Liquiritin apioside (0.1 mM) overnight, and capsaicin-induced currents are measured by patch-clamp electrophysiology. Current amplitude, rise time, and decay time are analyzed. The percentage of capsaicin-responsive neurons is determined.
|
| Animal Protocol |
In vivo animal studies for Liquiritin apioside are performed using rat pups. Liquiritin apioside (10 μg/μL) is administered intralaryngeally or perineurally as a single dose. The laryngeal chemoreflex is induced by capsaicin, hydrochloric acid, or distilled water. Apnea, hypertension, and bradycardia responses are measured at 10 and 20 minutes post-administration. The laryngeal mechanoreflex is also assessed to confirm selectivity.

| ADME/Pharmacokinetics |
Liquiritin apioside has a molecular weight of 550.51 and molecular formula C26H30O13. It is soluble in DMSO (100 mg/mL, 181.65 mM). For in vivo studies, formulations include 10% DMSO + 40% PEG300 + 5% Tween-80 + 45% saline (≥2.5 mg/mL). The compound should be stored at 4°C protected from light; in solvent at -80°C for 6 months or at -20°C for 1 month (protected from light).
|
| Toxicity/Toxicokinetics |
Toxicological data for Liquiritin apioside are limited. The compound is a natural flavonoid from licorice and is generally considered to have low toxicity. Standard laboratory safety precautions should be followed when handling the compound. No specific acute toxicity data have been reported in the literature. The compound is for research use only and is not for human therapeutic use.

| Additional Infomation |
Glycyrrhizin is a flavonoid compound belonging to the glycoside class. It has been reported to exist in licorice (Glycyrrhiza uralensis), wild licorice (Glycyrrhiza aspera), and other organisms with relevant data. See also: Licorice (Glycyrrhiza uralensis) root (part).
Liquiritin apioside is a flavonoid compound found in licorice (Glycyrrhiza uralensis) roots. It is one of the main active components in Suan-Zao-Ren decoction, a traditional Chinese medicine used for treating insomnia. The compound is an orally active TRPV1 receptor inhibitor that selectively inhibits laryngeal chemoreflex (LCR) without affecting mechanoreflex (LMR). It has anti-tussive properties and can be used in research of respiratory-related diseases. Liquiritin apioside modulates oxidative stress, regulates neurotransmitter balance, and suppresses pro-inflammatory mediators. It is for research use only and has not been approved for clinical use. |
