| Targets |
Lathosterol does not have a traditional receptor target. It functions as a key intermediate in the cholesterol biosynthesis pathway. It is a substrate for the enzyme lathosterol oxidase (also known as sterol-C5-desaturase), which catalyzes its conversion to 7-dehydrocholesterol. Its levels reflect the activity of the cholesterol synthesis pathway.
|
|---|---|
| ln Vitro |
In vitro, lathosterol is used as a biomarker to study cholesterol metabolism. Its levels can be measured in cell culture to assess cholesterol synthesis. It is a substrate for enzymes in the cholesterol biosynthesis pathway. Its role as a metabolite is well-established.
|
| ln Vivo |
In vivo, lathosterol is a clinically relevant biomarker for cholesterol synthesis. Elevated serum levels indicate increased endogenous cholesterol production. It is measured to assess the effectiveness of statin therapy and to study cholesterol metabolism in various disease states.
|
| Enzyme Assay |
In vitro enzyme assays for lathosterol involve measuring its conversion to 7-dehydrocholesterol by the enzyme lathosterol oxidase. The reaction can be monitored by HPLC or LC-MS. These assays are used to study cholesterol biosynthesis and the effects of inhibitors on this pathway.
|
| Cell Assay |
In vitro cell-based assays for lathosterol involve treating cells with compounds that affect cholesterol synthesis and then measuring intracellular lathosterol levels by LC-MS. This is used to assess the activity of the cholesterol biosynthesis pathway.
|
| Animal Protocol |
In vivo animal experiments for lathosterol are not typically performed. Its role as a clinical biomarker is well-established. Animal models of hypercholesterolemia can be used to study the relationship between lathosterol levels and cholesterol synthesis.
|
| ADME/Pharmacokinetics |
Lathosterol has a molecular weight of 386.65 and a molecular formula of C27H46O. It has a density of 0.98 g/cm3 and a boiling point of 479.4°C. It is a white to off-white solid powder. It is soluble in DMSO.
|
| Toxicity/Toxicokinetics |
Lathosterol is an endogenous metabolite and is not considered toxic. It is a research chemical and a diagnostic marker. Standard safety precautions should be taken when handling. It is not intended for human use.

| Additional Infomation |
5α-Cholesterol-7-en-3β-ol is a cholesterol compound formed by the substitution of a β-hydroxyl group at the 3-position of 5α-cholesterol-7-ene. It is a metabolite in both humans and mice. It is a 3β-sterol, a cholesterol compound, a C27-steroid, and a Δ7-sterol. Lanosterol has been reported in Dictyuchus monosporus, Asterias amurensis, and other organisms with available data.
Lathosterol is an intermediate in cholesterol biosynthesis. It is formed during the conversion of lanosterol to cholesterol. Serum lathosterol is a biomarker for systemic cholesterol synthesis. It is a human metabolite. It is used as a research tool to study cholesterol metabolism. |

| Solubility (In Vivo) |
Solubility in Formulation 1: ≥ 2 mg/mL (5.17 mM) (saturation unknown) in 10% EtOH + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.0 mg/mL clear EtOH stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution. Solubility in Formulation 2: ≥ 2 mg/mL (5.17 mM) (saturation unknown) in 10% EtOH + 90% Corn Oil (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.0 mg/mL clear EtOH stock solution to 900 μL of corn oil and mix evenly.  (Please use freshly prepared in vivo formulations for optimal results.) |

Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
