| Targets |
Fluorescein-5-maleimide targets thiol groups (cysteine residues) on proteins and peptides. The maleimide group reacts specifically with sulfhydryl groups under mild conditions, forming a stable thioether bond. This allows for site-specific fluorescent labeling of proteins for detection and imaging applications.
|
|---|---|
| ln Vitro |
In vitro, Fluorescein-5-maleimide is used as a fluorescent labeling reagent for proteins and peptides. It covalently reacts with thiol groups on cysteine residues, enabling the detection and visualization of labeled proteins in various assays. Its excitation wavelength is 494 nm and emission wavelength is 519 nm, making it suitable for standard fluorescein detection channels.
|
| ln Vivo |
In vivo, Fluorescein-5-maleimide is not administered as a therapeutic agent. It may be used to label proteins or peptides that are subsequently administered to animals for imaging studies, allowing for the tracking of labeled molecules in vivo.
|
| Enzyme Assay |
In vitro labeling assays with Fluorescein-5-maleimide are conducted by incubating the dye with protein or peptide samples containing free thiol groups. The reaction is typically performed in pH 6.5-7.5 buffer at room temperature for 1-2 hours. Unreacted dye is removed by desalting or dialysis. Labeled proteins are analyzed by SDS-PAGE with fluorescence detection or by spectrophotometry.
|
| Cell Assay |
Fluorescein-5-maleimide is used in cellular labeling assays to tag proteins of interest. Cells are incubated with the dye or with dye-labeled proteins. Fluorescence is detected using flow cytometry or fluorescence microscopy (excitation 494 nm, emission 519 nm). The dye can be used to study protein localization, trafficking, and interactions.
|
| Animal Protocol |
In vivo animal studies with Fluorescein-5-maleimide involve the administration of dye-labeled proteins or peptides to animals for imaging applications. The labeled molecules are injected intravenously or intraperitoneally. Fluorescence imaging is performed using appropriate excitation/emission filters to track the biodistribution and accumulation of the labeled molecules.
|
| ADME/Pharmacokinetics |
Fluorescein-5-maleimide is a fluorescent dye and does not have pharmacokinetic properties as a drug. It is applied exogenously in research settings.
|
| Toxicity/Toxicokinetics |
Fluorescein-5-maleimide is a fluorescent dye and is not intended for human consumption. Standard laboratory safety precautions should be followed.
|

| Solubility (In Vivo) |
Solubility in Formulation 1: 2.5 mg/mL (5.85 mM) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), suspension solution; with sonication.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution. Solubility in Formulation 2: 2.08 mg/mL (4.87 mM) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), suspension solution; with ultrasonication. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 20.8 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL. Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.  (Please use freshly prepared in vivo formulations for optimal results.) |

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
